Identification of a Kupffer Cell Subset Capable of Reverting the T Cell Dysfunction Induced by Hepatocellular Priming

Researchers used a mouse model of hepatitis B virus (HBV) infection, in which HBV-specific naive CD8+ T cells recognizing hepatocellular antigens were driven into a state of immune dysfunction, to identify a subset of Kupffer cells that cross-presented hepatocellular antigens upon interleukin-2 administration.

TAZ Is Indispensable for C-MYC-Induced Hepatocarcinogenesis

Scientists investigated the functional contribution of Yes-associated protein and/or transcriptional co-activator with PDZ-binding motif (TAZ) during c-MYC induced liver tumor development.

Therapeutic HNF4A mRNA Attenuates Liver Fibrosis in a Preclinical Model

Researchers investigated restoration of hepatocyte functions by HNF4A mRNA transfection in vitro, and analyzed the attenuation of liver fibrosis and cirrhosis in multiple mouse models, by delivering hepatocyte-targeted biodegradable lipid nanoparticles encapsulating HNF4A mRNA.

The Anti-Fibrotic Drug Pirfenidone Inhibits Liver Fibrosis by Targeting the Small Oxidoreductase Glutaredoxin-1

The authors showed that pirfenidone, an anti-lung fibrosis drug, inhibited hepatic stellate cell activation and liver fibrosis in a Glutaredoxin-1 (GLRX)-dependent manner. Glrx depletion exacerbated liver fibrosis, and decreased GLRX and increased protein S-glutathionylation were observed in fibrotic mouse and human livers.

    AAV Integration in Human Hepatocytes

Investigators ascertained the integration frequency of recombinant adeno-associated viral (AAV) vectors in human hepatocytes transduced either ex vivo or in vivo and subsequently expanded in a mouse model of xenogeneic liver regeneration.

Single Cell and Plasma RNA Sequencing for RNA Liquid Biopsy for Hepatocellular Carcinoma

Single-cell RNA sequencing was performed to decipher transcriptomic profiles of single cells from hepatocellular carcinoma (HCC) samples. Cell-type-specific transcripts were identified and used for deducing the cell-type-specific gene signature scores of plasma RNA from patients with and without HCC.

Thrombopoietin from Hepatocytes Promotes Hematopoietic Stem Cell Regeneration after Myeloablation

Using a Thpo translational reporter, scientists found that the liver but not the bone marrow was the major source of thrombopoietin protein after myeloablation. Mice with conditional Thpo deletion from hepatocytes showed significant defects in hematopoietic stem cell regeneration and hematopoietic rebound after myeloablation.

    Abstract

Human Induced Pluripotent Stem Cell-Derived Macrophages Ameliorate Liver Fibrosis

Investigators demonstrated that treatment of liver fibrosis with both human induced pluripotent stem cell-derived macrophage populations and especially M2 subtype significantly reduced fibrogenic gene expression and disease associated histological markers in immunodeficient Rag2−/−γc−/− mice model, making this approach a promising cell-based avenue to ameliorate fibrosis.

Pantoprazole Ameliorates Liver Fibrosis and Suppresses Hepatic Stellate Cell Activation in Bile Duct Ligation Rats by Promoting YAP Degradation

Researchers investigated the effects of pantoprazole (PPZ), a proton pump inhibitor, against liver fibrosis in a bile duct ligation rat model, human hepatic stellate cell (HSC) line LX-2 and mouse primary HSCs, and explored the potential mechanisms underlying the effects of PPZ in vitro and in vivo.

PPM1D Is a Potential Prognostic Biomarker and Correlates with Immune Cell Infiltration in Hepatocellular Carcinoma

Protein phosphatase magnesium-dependent 1 delta (PPM1D) mRNA expression was significantly higher in several human cancers, including hepatocellular carcinoma (HCC), than in the corresponding normal human tissues. The PPM1D mRNA high expression in HCC was significantly correlated with poor prognosis.

Exosomal microRNA-15a from Mesenchymal Stem Cells Impedes Hepatocellular Carcinoma Progression via Downregulation of SALL4

The authors explored the role of exosomal miR-15a derived from mesenchymal stem cells (MSCs) and its related mechanisms in hepatocellular carcinoma (HCC).The exosomes from MSCs restrained HCC cell proliferative, migrating, and invasive potentials, and accelerated their apoptosis.

Deletion of RAGE Fails to Prevent Hepatosteatosis in Obese Mice Due to Impairment of Other AGEs Receptors and Detoxifying Systems

Scientists analyzed the effect of obesity on advanced glycation endproducts (AGEs) accumulation, AGE-receptors and AGE-detoxification, and whether the absence of RAGE may have improved hepatosteatosis and inflammation, by comparing the liver of lean control, obese and obese RAGE-deficient mice.
