Increased Expression or Activation of TRPML1 Reduces Hepatic Storage of Toxic Z Alpha-1 Antitrypsin

The authors showed that by increasing lysosomal exocytosis, transient receptor potential mucolipin 1 (TRPML1) gene transfer or small molecule-mediated activation of TRPML1 reduced hepatic mutant Z-alpha-1 antitrypsin (ATZ) globules and fibrosis in PiZ transgenic mice that expressed the human ATZ.

LY6D Is Crucial for Lipid Accumulation and Inflammation in Nonalcoholic Fatty Liver Disease

Investigators fed C57BL6/J mice one of three diets, a standard chow diet, high-sucrose diet, or high-fat diet, and then characterized them. The mice fed a high-sucrose diet had more severely compacted macrovesicular and microvesicular lipid droplets than those in the other groups.

Ablation of the Deubiquitinase USP15 Ameliorates Nonalcoholic Fatty Liver Disease and Nonalcoholic Steatohepatitis

Researchers observed that deubiquitinase USP15 expression was upregulated in the livers of mice fed a high-fat diet and liver biopsies of patients with nonalcoholic fatty liver disease or nonalcoholic steatohepatitis.

Fatty Acid Metabolization and Insulin Regulation Prevent Liver Injury from Lipid Accumulation in Himalayan Marmots

The authors explored the lipid accumulation and metabolic processes of the Himalayan marmot, a hibernating rodent species. They found that the unsaturated fatty acid (UFA) content in food was consistent with a large increase in the body mass of Himalayan marmots.

KLF6 Alleviates Hepatic Ischemia-Reperfusion Injury by Inhibiting Autophagy

After ischemia-reperfusion (I/R) injury, scientists found that Krüppel-like factor (KLF) 6 expression in mice and hepatocytes was significantly upregulated. Mice were then subjected to I/R following injection of shKLF6- and KLF6-overexpressing adenovirus through the tail vein.

UHRF1 Inhibition Epigenetically Reprograms Cancer Stem Cells to Suppress the Tumorigenic Phenotype of Hepatocellular Carcinoma

Researchers investigated the role and mechanism of ubiquitin-like with PHD and ring finger domains 1 (UHRF1) in regulating cancer stem cell properties and evaluated the impact of UHRF1 targeting on HCC.

6-Methyl Flavone Inhibits Nogo-B Expression and Improves High Fructose Diet-Induced Liver Injury in Mice

Investigators tested 14 flavones/isoflavones in hepatocytes using dual luciferase reporter system based on the Nogo-B transcriptional response system, and found that 6-methyl flavone exerted the strongest inhibition on Nogo-B expression in hepatocytes.

The Elk-3 Target Abhd10 Ameliorates Hepatotoxic Injury and Fibrosis in Alcoholic Liver Disease

Analyses of liver tissue samples from severe alcoholic hepatitis patients and two murine models of alcoholic liver disease (ALD) revealed that the ALD phenotype was associated with upregulation of ETS domain-containing protein (ELK-3) and ELK-3 signaling activity.

Optimization of Physical Microenvironment to Maintain the Quiescence of Human Induced Pluripotent Stem Cell-Derived Hepatic Stellate Cells

Scientists generated quiescent-like hepatic stellate cells (iqHSCs) from human induced pluripotent stem cells and developed a culture method to maintain such iqHSCs in a lowly activated state for up to five days by optimizing their physical culture microenvironment.

Lifestyle Interventions in Nonalcoholic Fatty Liver Disease

Lifestyle interventions, including diet and physical activity, are the main treatments for nonalcoholic fatty liver disease. The Mediterranean diet, or a diet similar in its culturally sensitive components, is the most effective diet for losing weight.
