TOP STORYUsing a model of spontaneous autoimmune arthritis, scientists showed that arthritogenic Th17 cells stimulated fibroblast-like synoviocytes via interleukin-17 to secrete the cytokine GM-CSF and also expanded synovial-resident innate lymphoid cells in inflamed joints. PUBLICATIONS(Ranked by impact factor of the journal)With novel, immunocompetent metastasis models, researchers demonstrated that tumor cell derived CCL2-mediated recruitment of inflammatory monocytes (IMs) is necessary and sufficient for lung squamous carcinomas (LUSC) metastasis. They showed that IMs highly express Factor XIIIA, which promoted fibrin cross-linking to create a scaffold for LUSC cell invasion and metastases. [Nat Commun] Full Article Investigators showed that B cell-specific deletion of the myosin IIa heavy chain reduced the numbers of bone marrow B cell precursors and splenic marginal zone, peritoneal B1b, and germinal center B cells. Myosin IIa-deficient follicular B cells acquired an activated phenotype and were less efficient in chemokinesis and extraction of membrane-presented antigens. [Cell Rep] Full Article | Graphical Abstract Regulation of Pathogenic T Helper 17 Cell Differentiation by Steroid Receptor Coactivator-3 Scientists showed that steroid receptor coactivator-3 (SRC-3) critically regulates T helper 17 (Th17) cell differentiation. Reduced incidence of experimental autoimmune encephalitis associated with decreased Th17 cell generation in vivo was observed in mice with SRC-3 deletion specifically in T cells. [Cell Rep] Full Article | Graphical Abstract IL-37 Isoform D Downregulates Pro-Inflammatory Cytokines Expression in a Smad3-Dependent Manner Investigators showed that IL-37d possesses anti-inflammatory roles both in vitro and in vivo. IL-37d interacted with Smad3 and promoted nuclear translocation of pSmad3. [Cell Death Dis] Full Article Researchers devised a combination therapy of peptide/MHC tetramers and IL-2/anti-IL-2 monoclonal antibody complexes to generate antigen-specific Tregs and maintain them over extended time periods. [Sci Rep] Full Article Scientists showed that specific immunotherapy maintained airway epithelial homeostasis in mice exposed to dermatophagoides farinae, which induced increased expression of IL-25, endoplasmic reticulum stress and airway epithelial apoptosis. [Sci Rep] Full Article Investigators showed that autoimmune regulator (AIRE) finds differential expression in androgen-dependent and -independent prostate cancer cells. AIRE expression was more in androgen-independent cells due to its regulation by transcription factor Elk-1. [Oncogenesis] Full Article Scientists showed that gastric cancer (GC)-derived exosomes induced programmed cell death 1 (PD1)+ tumor-associated macrophage generation, and produced a large number of IL-10, impaired CD8+ T-cell function, and created conditions that promote GC progression. [Oncogenesis] Full Article The authors successfully extracted exosomes from Pseudomonas aeruginosa and assessed the effect of those exosomes on the development of the allergic response in two types of classic asthma models. REVIEWSDefining and Understanding Adaptive Resistance in Cancer Immunotherapy The authors categorize three types of resistance, namely target-missing, primary, and acquired resistance. This categorization requires reliable, accurate tissue sampling and appropriate interpretation of results based on the four classifications of tumor immunity in the microenvironment. POLICY NEWSItaly’s New Prime Minister Defended Discredited Stem Cell Therapy The electoral victory of two populist parties in March had Italian scientists worried that a new era of antiscientific policies might be around the corner—and yesterday’s appointment of Giuseppe Conte as the country’s new prime minister is making some even more nervous. [ScienceInsider] Editorial Swedish Court Blocks New Home for Nobel Foundation A Swedish court has blocked construction of a controversial new Nobel Center planned for central Stockholm’s waterfront. The eight-story, brass-clad structure is expected to serve as a hub for the Nobel Foundation’s activities, including the annual December award ceremonies for the world’s most prestigious science prizes. [ScienceInsider] Editorial Scientist Who Received Millions from NIH Leaves Alabama Posts A professor who received about $5 million in grants from the National Institutes of Health left his posts at the University of Alabama, Birmingham (UAB), and Birmingham VA Medical Center last year after an investigation found evidence of image manipulation in 20 of his papers, Retraction Watch has learned.
