Meningeal Lymphatics Affect Microglia Responses and Anti-Aβ Immunotherapy

Investigators showed that ablation of meningeal lymphatic vessels in 5xFAD mice (a mouse model of amyloid deposition that expressed five mutations found in familial Alzheimer’s disease) worsened the outcome of mice treated with anti-Aβ passive immunotherapy.

Cancer Cells Retrace a Stepwise Differentiation Program during Malignant Progression

Using multi-omics approaches, researchers revealed that metastasis-like primary tumors arose from IT via dedifferentiation following a reverse trajectory along the developmental pathway of islet B-cells, which resulted in the acquisition of a progenitor-like molecular phenotype.

    Abstract

Adrenergic Regulation of the Vasculature Impairs Leukocyte Interstitial Migration and Suppresses Immune Responses

The authors investigated how adrenoceptor signaling influenced leukocyte behavior. Intravital two-photon imaging after injection of noradrenaline revealed transient inhibition of CD8+ and CD4+ T cell locomotion in tissues.

Optogenetic Activation of Local Colonic Sympathetic Innervations Attenuates Colitis by Limiting Immune Cell Extravasation

Scientists investigated the impact of the local arm of the sympathetic nervous system in an inflammatory response in the colon. Intra-rectal insertion of an optogenetic probe in mice engineered to express channelrhodopsin-2 in tyrosine hydroxylase cells activated colonic sympathetic fibers.

Tumors Exploit FTO-Mediated Regulation of Glycolytic Metabolism to Evade Immune Surveillance

Researchers identified the m6A demethylase FTO as an essential epitranscriptomic regulator utilized by tumors to escape immune surveillance through regulation of glycolytic metabolism.

Gremlin 1+ Fibroblastic Niche Maintains Dendritic Cell Homeostasis in Lymphoid Tissue

Investigators used single-cell RNA sequencing of human and mouse lymph nodes to identify a subset of T cell–zone fibroblastic reticular cells defined by the expression of Gremlin1 in both species.

    Abstract

Heme Catabolism by Tumor-Associated Macrophages Controls Metastasis Formation

Scientists showed that a distinct subset of tumor-associated macrophages, endowed with high rates of heme catabolism by the stress-responsive enzyme heme oxygenase-1, played a critical role in shaping a prometastatic tumor microenvironment favoring immunosuppression, angiogenesis and epithelial-to-mesenchymal transition.

    Abstract

Dietary Conjugated Linoleic Acid Links Reduced Intestinal Inflammation to Amelioration of CNS Autoimmunity

The authors report that dietary supplementation with conjugated linoleic acid, a mixture of linoleic acid isomers, ameliorated CNS autoimmunity in a spontaneous mouse model of multiple sclerosis, accompanied by an attenuation of intestinal barrier dysfunction and inflammation as well as an increase in intestinal myeloid-derived suppressor-like cells.

Bruton’s Tyrosine Kinase Regulates Gut Immune Homeostasis through Attenuating Th1 Response

Researchers showed that Bruton’s tyrosine kinase-deficiency promoted inflammatory bowel disease progression in a mouse model of colitis. The increased colitis susceptibility of BTK-deficient mice was not caused by gut microbiota changes but rather arose from enhanced pro-inflammatory Th1 response.

    Therapeutic Cancer Vaccines

Scientists propose strategies for combining suitable vaccine platforms with novel immunomodulatory approaches and standard-of-care treatments for overcoming tumor resistance and enhancing clinical efficacy.

Fibroblasts as Immune Regulators in Infection, Inflammation and Cancer

The authors compare the mechanisms by which fibroblasts control local immune responses, as well as the factors regulating their inflammatory and suppressive profiles, in different tissues and pathological settings.

Epithelial–Immune Cell Interplay in Primary Sjögren Syndrome Salivary Gland Pathogenesis

Researchers discuss changes in the cells of the salivary gland epithelium in primary Sjögren syndrome, and the proposed interplay of these cells with environmental stimuli and the immune system.
