The Cholesterol Metabolite 25-Hydroxycholesterol Restrains the Transcriptional Regulator SREBP2 and Limits Intestinal IgA Plasma Cell Differentiation

Researchers examined how the cholesterol metabolite 25-hydroxycholesterol (25-HC) impacted the intestinal B cell response. Mice lacking cholesterol 25-hydroxylase, the enzyme generating 25-HC, had higher frequencies of immunoglobulin A-secreting antigen-specific B cells upon immunization or infection.

IL-33-Induced Metabolic Reprogramming Controls the Differentiation of Alternatively Activated Macrophages and the Resolution of Inflammation

Scientists showed that uncoupling protein-2-mediated mitochondrial reprogramming and the transcription factor GATA3 specifically controlled the differentiation of pro-resolving alternatively activated macrophages in response to the alarmin IL-33.

Dynamic Changes in tRNA Modifications and Abundance during T Cell Activation

Investigators examined the transfer RNA (tRNA) and mRNA pools of T cells during their vigorous proliferation and differentiation upon triggering their antigen receptor.

PI3Kδ Coordinates Transcriptional, Chromatin, and Metabolic Changes to Promote Effector CD8+ T Cells at the Expense of Central Memory

Using patient samples and a mouse model, the authors demonstrated that, upon activation, Pik3cdE1020K/+ CD8+ T cells exhibited exaggerated features of effector populations both in vitro and after viral infection that was associated with increased Fas-mediated apoptosis.

Proinflammatory Cytokines Promote TET2-Mediated DNA Demethylation during CD8 T Cell Effector Differentiation

To gain insight into the signaling determinants of effector-associated DNA methylation programming among CD8 T cells, scientists explored the role of interleukin-12 in the imprinting of IFNg expression during CD8 T cell priming.

Alloantigen-Activated (AAA) CD4+ T Cells Reinvigorate Host Endogenous T Cell Immunity to Eliminate Pre-Established Tumors in Mice

Alloantigen-activated CD4+ T cells were generated from CD4+ T cells isolated from BALB/c mice in cultures with dendritic cells induced from C57BL/6 mice.

Adoptive CD8T Cell Grafted with Liposomal Immunotherapy Drugs to Counteract the Immune Suppressive Tumor Microenvironment and Enhance Therapy for Melanoma

The immunosuppressive tumor microenvironment has become a formidable obstacle to the treatment of tumors using adoptive T cell therapy, in particular solid tumors. Effector OT-1 CD8+ T cells conjugated with liposomal immune regulators were developed to address this issue.

RSV Infection-Elicited High MMP-12–Producing Macrophages Exacerbate Allergic Airway Inflammation with Neutrophil Infiltration

Investigators showed that respiratory syncytial virus (RSV)-induced alveolar macrophages, which produced high levels of matrix metalloproteinase-12, exacerbated allergic airway inflammation with increased neutrophil infiltration.

Estrogen Deprivation Induces Chemokine Production and Immune Cell Recruitment in In Vitro and In Vivo Models of Estrogen Receptor-Positive Breast Cancer

Researchers investigated the effect of estrogen deprivation on the expression of chemokines and immune infiltration in vitro and in an ER+ immunocompetent mouse model.
