Epithelial Nlrp10 Inflammasome Mediates Protection against Intestinal Autoinflammation

In vivo, Nlrp10 signaling was dispensable during steady state but became functional during autoinflammation in antagonizing mucosal damage.

The Transcription Factor Mef2d Regulates B:T Synapse–Dependent GC-TFH Differentiation and IL-21–Mediated Humoral Immunity

Scientists found that myocyte enhancer factor 2 d (Mef2d) regulated Sh2d1a expression via DNA binding–dependent transcriptional repression, inhibiting SAP-dependent B:T synapse formation, and preventing antigen-specific CD4 T cells from differentiating into germinal center–T follicular helper (GC-TFH) cells.

IL-6 in the Infarcted Heart Is Preferentially Formed by Fibroblasts and Is Modulated by Purinergic Signaling

In order to understand which cardiac cell type preferentially contributes to IL-6 and how its production is regulated, investigators studied the cellular source and purinergic regulation of IL-6 formation in a murine myocardial infarction model.

Aurora A Kinase Inhibition Elevates PD-L1 Expression and Compromises Its Anti-Tumor Efficacy

The upregulation of the checkpoint protein PD-L1 reduced antitumor immunity in immune-competent mice, paradoxically inhibiting the antitumor effects of alisertib.

    Abstract

VMP1 Prevents Ca2+ Overload in Endoplasmic Reticulum and Maintains Naive T Cell Survival

VMP1 promoted Ca2+ release from endoplasmic reticulum (ER) under steady state, and its deficiency led to ER Ca2+ overload, ER stress, and secondary Ca2+ overload in mitochondria, resulting in massive apoptosis of naive T cells and defective T cell response.

    Abstract

Identification of IgA Autoantibodies Targeting Mesangial Cells Redefines the Pathogenesis of IgA Nephropathy

As in patients with immunoglobulin A (IgA) nephropathy (IgAN) , IgA+ plasmablasts accumulated in the kidneys of a murine model. IgA antibodies cloned from the plasmablasts carried substantial V-region mutations and bound to βII-spectrin and the surface of mesangial cells.

Molecular and Cognitive Signatures of Aging Partially Restored through Synthetic Delivery of IL2 to the Brain

The authors tested a brain-specific IL2 delivery system for the prevention of neurological decline in aging mice. They identified the molecular hallmarks of aging in the brain glial compartments and identified partial restoration of this signature through IL2 treatment.

Deubiquitinase OTUD6A in Macrophages Promotes Intestinal Inflammation and Colitis via Deubiquitination of NLRP3

Researchers investigated the expression and role of ovarian tumor deubiquitinase 6 A (OTUD6A) in inflammatory bowel disease. Bone marrow-derived macrophages were isolated from wild-type and Otud6a−/− mice to dissect molecular mechanisms.

NF-κB Activator 1 Downregulation in Macrophages Activates STAT3 To Promote Adenoma-Adenocarcinoma Transition and Immunosuppression in Colorectal Cancer

The authors investigated the effect of the NF-κB activator 1 downregulation of macrophages in the adenoma-adenocarcinoma transition using murine models, histological analysis, primary cell isolation, a co-culture system, and other methods.

Neuroimmune Mechanisms and Therapies Mediating Post-Ischaemic Brain Injury and Repair

Scientists review neuroimmune mechanisms in the context of post-ischaemic brain injury and repair, inlcuding how inflammatory immune cells exacerbate ischaemic neuronal injury after the onset of brain ischaemia, yet some of the immune cells thereafter change their function to neural repair.

Involvement of the Secosteroid Vitamin D in Autoimmune Rheumatic Diseases and COVID-19

The authors review the latest scientific and clinical evidence concerning vitamin D and immune response in autoimmune rheumatic diseases and COVID-19, which justify the need for monitoring of serum 25(OH)D3 concentrations and for appropriate supplementation following clinical trial-based approaches.

FDA Floats Oncology Authorization Shake-Up, Pitching ‘One-Trial’ Model for Accelerated and Full Approval

In draft guidance, the FDA outlined plans to move away from single-arm studies and instead favor randomized controlled trials that can potentially serve as the basis for both accelerated and full approval.
