Reciprocal Inhibition Between TP63 and STAT1 Regulates Anti-Tumor Immune Response through Interferon-γ Signaling in Squamous Cancer

Researchers identified that the squamous cell carcinomas-master transcription factor TP63 suppressed interferon-γ signaling. TP63 inhibition led to increased CD8⁺ T cell infiltration and heightened tumor killing in in vivo syngeneic mouse model and ex vivo co-culture system, respectively.

ATG-101 Is a Tetravalent PD-L1×4–1BB Bispecific Antibody that Stimulates Anti-Tumor Immunity through PD-L1 Blockade and PD-L1-Directed 4–1BB Activation

To effectively target 4-1BB and treat diseases resistant to immune checkpoint inhibitors, scientists engineered ATG-101, a tetravalent “2+2” PD-L1×4-1BB bispecific antibody. ATG-101 bound PD-L1 and 4-1BB concurrently, with a greater affinity for PD-L1, and potently activated 4-1BB⁺ T cells.

Tonic TCR and IL-1β Signaling Mediate Phenotypic Alterations of Naive CD4⁺ T Cells

The authors identified that interleukin (IL)-1β signaling affects Inert naive CD4⁺ T (TN) cell characteristics, independent of tonic T cell receptor signaling, which also altered TN cell phenotypes. IL-1β reversibly attenuates the differentiation potential of TN cells toward Treg cells.

Scientists investigated the impact of enterovirus 71 (EV71) on plasmacytoid dendritic cells (pDCs) and CD4⁺ T cells. The results showed that pDCs were promptly activated, secreting interferon (IFN)-α and inducing CD4⁺ T cell proliferation and differentiation during early EV71 infection.
