SCIENCE NEWS Dermatology: Watching Immune Cell Movement To and From the Skin A team of researchers has now used mice engineered to express the photoconvertible fluorescence protein Kaede, which changes from green to red when exposed to violet light, to track regulatory T cell movement under physiologic conditions and during immune responses in the skin. CURRENT PUBLICATIONS In Vivo Requirement for Atg5 in Antigen Presentation by Dendritic Cells Mice with dendritic cell-conditional deletion in Atg5, a key autophagy gene, showed impaired CD4+ T cell priming after herpes simplex virus infection and succumbed to rapid disease. [Immunity] Activated Regulatory T Cells are the Major T Cell Type Emigrating from the Skin During a Cutaneous Immune Response In Mice Results suggest that regulatory T cells in circulation infiltrate the periphery, traffic to draining lymph nodes, and then recirculate back to the skin, contributing to the downregulation of cutaneous immune responses. [J Clin Invest] Competing Feedback Loops Shape IL-2 Signaling Between Helper and Regulatory T Lymphocytes in Cellular Microenvironments Researchers combined mathematical modeling and experiments to dissect the dynamics of the interleukin-2 (IL-2) signaling network that links the prototypical IL-2 producers, conventional T helper cells, and regulatory T cells. [Proc Natl Acad Sci USA] Expression of Helios, an Ikaros Transcription Factor Family Member, Differentiates Thymic-Derived from Peripherally Induced Foxp3+ T Regulatory Cells Helios, a member of the Ikaros transcription factor family, is preferentially expressed at the mRNA level by regulatory T cells. Researchers evaluated Helios protein expression using a newly generated monoclonal antibody and demonstrated that it is expressed in all thymocytes at the double negative 2 stage of thymic development. [J Immunol] A Regulatory Role for Macrophage Class A Scavenger Receptors in TLR4-Mediated LPS Responses Researchers have revealed a mechanism where scavenger receptors regulate the activation status of the immune system and are involved in balancing a proper immune response to infection. This regulation could also be important in maintaining tolerance since these receptors have been shown to be involved in regulation of self reactivity. [Eur J Immunol] Foxp3-Transduced Polyclonal Regulatory T Cells Suppress Natural Killer Cell Functions in a TGF-Beta Dependent Manner The purpose of this study was to investigate the suppressive effect of Foxp3-transduced CD4+ T cells on natural killer (NK) cell functions, and thus to present a possible method for managing NK cell-related autoimmune abnormalities. [Autoimmunity] IL-10 is Required for Human CD4+CD25+ Regulatory T Cell-Mediated Suppression of Xenogeneic Proliferation In this study, researchers analyzed whether interleukin-10 (IL-10) is required for regulatory T cells to suppress xenogeneic responses in vitro by small interfering RNA-mediated IL-10 knockdown. [Immunol Cell Biol] Elevated Expression of Foxp3 in Tumor-Infiltrating Regulatory T Cells Suppresses T Cell Proliferation and Contributes to Gastric Cancer Progression in a COX-2-Dependent Manner Data demonstrate a mechanism by which tumor-infiltrating regulatory T cells with increased Foxp3 expression can mediate immune suppression via COX-2/PGE2 production in the gastric cancer microenvironment.
