    TOP STORY

Researchers Discover New Signaling Pathway Linked to Inflammatory Disease  ShareThis
Scientists have described for the first time a key inhibitory role for the IL-1 signaling pathway in the human innate immune system, providing novel insights into human inflammatory bowel disease and potential new treatments. [Press release from the University of California, San Diego discussing online prepublication in The Journal of Experimental Medicine]

    SCIENCE NEWS

Yale Researchers Find A Surprising Culprit in Lupus
Yale University researchers were able to reduce symptoms of lupus in mice by eliminating a key immune system cell and in doing so may have identified a new therapeutic target for a variety of other autoimmune diseases. [Press release from Yale University discussing online prepublication in Immunity]

Statin Use Linked to Rare Autoimmune Muscle Disease
Researchers have discovered how statins, the most commonly prescribed class of medication in the United States, appear to trigger a rare but serious autoimmune muscle disease in a small portion of the 30 million Americans who take the cholesterol-lowering drugs. [Press release from Johns Hopkins Medicine discussing online prepublication in Arthritis & Rheumatism]

Zebrafish Provide New Hope for Cancer Treatment
The imaging of tumor growth in zebrafish has revealed for the first time how cancer cells have the capacity to co-opt the immune system into spreading disease, leading the way for investigations into potential therapies for eliminating early-stage cancer in humans. [Press release from the University of Bristol discussing online prepublication in PLoS Biology]

UCSF “Fountain of Youth” Pill Could Restore Aging Immune System
Researchers have identified an existing medication that restores key elements of the immune system that, when out of balance, lead to a steady decline in immunity and health as people age. [Press release from the University of California, San Francisco discussing online prepublication in Clinical Immunology]

CURRENT PUBLICATIONS (Ranked by Impact Factor of the Journal)

T-Bet Represses TH17 Differentiation by Preventing Runx1-Mediated Activation of the Gene Encoding ROR(Gamma)t
Here, researchers report that the transcription factor T-bet suppressed development of the TH17 cell lineage by inhibiting transcription of Rorc (which encodes the transcription factor ROR(gamma)t). [Nat Immunol]

Autophagy Proteins Regulate Innate Immune Responses by Inhibiting the Release of Mitochondrial DNA Mediated by the NALP3 Inflammasome
Here, researchers demonstrate that depletion of the autophagic proteins LC3B and beclin 1 enhanced the activation of caspase-1 and secretion of interleukin 1beta (IL-1beta) and IL-18. [Nat Immunol]

Foxo Transcription Factors Control Regulatory T Cell Development and Function
Here, researchers showed the absence of Foxo1 severely curtailed the development of Foxp3+ regulatory T cells and those that developed were nonfunctional in vivo. [Immunity]

Dendritic Cells in Lupus Are Not Required for Activation of T and B Cells but Promote Their Expansion, Resulting in Tissue Damage
Dendritic cells (DCs) initiate and control the adaptive immune response against infections. However, their contributions to the anti-self adaptive immune response in autoimmune disorders like systemic lupus erythematosus are uncertain. By constitutively deleting DCs in MRL.Fas(lpr) mice, researchers show that they have complex roles in murine lupus. [Immunity]

Transcription Factor Miz-1 Is Required to Regulate Interleukin-7 Receptor Signaling at Early Commitment Stages of B Cell Differentiation
Researchers report that mice lacking the POZ (Poxvirus and zinc finger) domain of the transcription factor Miz-1 [Zbtb17(deltaPOZ/deltaPOZ) ] almost entirely lacked follicular B cells, as shown by the fact that their progenitors failed to activate the Jak-Stat5 pathway and to upregulate the antiapoptotic gene Bcl2 upon IL-7 stimulation. [Immunity]

A Peripheral CD4+ T Cell Precursor for Naive, Memory, and Regulatory T Cells
Researchers have previously shown that a subset of naive CD4+ T cells, defined by the expression on their surface of a very low density of CD44 [CD44(v.low) cells]. The data support a key role for CD4+ CD44(v.low) cells as peripheral precursors that maintain the integrity of the CD4+ T cell pool. [J Exp Med]

Autoantibodies Against 3-Hydroxy-3-Methylglutaryl-Coenzyme A Reductase (HMGCR) in Patients with Statin-Associated Autoimmune Myopathy
Statins up-regulate expression of HMGCR, the major target of autoantibodies in statin-associated immune-mediated necrotizing. Regenerating muscle cells express high levels of HMGCR, which may sustain the immune response even after statins are discontinued. [Arthritis Rheum]

Preferential Enhancement of Older Human T Cell Cytokine Generation, Chemotaxis, Proliferation and Survival by Lenalidomide
Lenalidomide enhanced proliferation and suppressed apoptosis of stimulated T cells from old subjects, by IL-2-dependent mechanisms, and restored diminished T cell chemotactic responses to CCL21 and sphingosine 1-phosphate. The reversal of T cell abnormalities of immunosenescence by low concentrations of lenalidomide suggest a potential for improvement of immunity in the elderly. [Clin Immunol]

Live Imaging of Innate Immune Cell Sensing of Transformed Cells in Zebrafish Larvae: Parallels between Tumor Initiation and Wound Inflammation
Here, researchers take advantage of the translucency of zebrafish larvae to observe the host innate immune cell response as oncogene-transformed melanoblasts and goblet cells multiply within the larval skin. [PLoS Biol]
