    TOP STORY
Scientists Shed New Light on Link between ‘Killer Cells’ and Diabetes
Killer T-cells in the human body which help protect us from disease can inadvertently destroy cells that produce insulin, new research has uncovered. [Press release from King’s College London discussing online prepublication in Nature Immunology] Press Release | Abstract

Rescued Tolerant CD8 T Cells Are Preprogrammed to Reestablish the Tolerant State
Using an in vivo mouse model, investigators show that tolerant T cells proliferate and become functional under lymphopenic conditions, even in a tolerogenic environment. [Science] Abstract

An Essential Role for TH2-Type Responses in Limiting Acute Tissue Damage during Experimental Helminth Infection
These studies indicate an essential role for TH2-type immune responses in mediating acute wound healing during helminth infection. [Nat Med] Abstract | Press Release

Cytometry by Time-of-Flight Shows Combinatorial Cytokine Expression and Virus-Specific Cell Niches within a Continuum of CD8+ T Cell Phenotypes
By using metal-labeled probes and mass spectrometric analysis (cytometry by time-of-flight) of human CD8+ T cells, researchers analyzed the expression of many more proteins than previously possible with fluorescent labels, including surface markers, cytokines, and antigen specificity with modified peptide-MHC tetramers. [Immunity] Abstract

The Transcription Factors T-bet and Eomes Control Key Checkpoints of Natural Killer Cell Maturation
Investigators show that mice lacking both transcription factors Eomesodermin (Eomes) and T-bet failed to develop natural killer cells. [Immunity] Abstract

Mycobacterium tuberculosis Inhibits Neutrophil Apoptosis, Leading to Delayed Activation of Naive CD4 T cells
Mycobacterium tuberculosis promotes its replication by inhibiting the apoptosis of infected macrophages. Researchers show that M. tuberculosis also suppresses neutrophil apoptosis. [Cell Host Microbe] Abstract

Dickkopf-3, an Immune Modulator in Peripheral CD8 T-Cell Tolerance
To determine the molecular basis of peripheral CD8 T-cell tolerance, researchers exploited a double transgenic mouse model in which CD8 T cells are neonatally tolerized following interaction with a parenchymal self-antigen. [Proc Natl Acad Sci USA] Abstract

CD4 and CD8 T Cells Require Different Membrane Gangliosides for Activation
While investigating T-cell activation in ganglioside-deficient mice, scientists observed that CD4+ and CD8+ T cells required different ganglioside subsets for activation. [Proc Natl Acad Sci USA] Abstract

Toll-Like Receptor 3 Signaling Converts Tumor-Supporting Myeloid Cells to Tumoricidal Effectors
Researchers find that the Toll-like receptor 3/Toll–IL-1 receptor domain-containing adaptor molecule 1 pathway is not only important to mature myeloid dendritic cells for cross-priming and natural killer cell activation in the induction of tumor immunity, but also critically engaged in tumor suppression by converting tumor-supporting macrophages to those with tumoricidal properties. [Proc Natl Acad Sci USA] Abstract

Morbid Obese Human Subjects Have Increased Peripheral Blood CD4+ T Cells with Skewing Toward a T-Regulatory- and T-Helper 2-Dominated Phenotype
In this study, researchers investigated the peripheral blood T-cell compartment of morbid obese and lean subjects. [Diabetes] Abstract

Tetracyclines Convert the Osteoclastic-Differentiation Pathway of Progenitor Cells to Produce Dendritic Cell-Like Cells
Osteoclasts and dendritic cells (DCs) are derived from common progenitors, such as bone marrow-derived macrophages. In this article, researchers show that tetracyclines convert the differentiation pathway, resulting in DC-like cells not osteoclasts. [J Immunol] Abstract

CD69 Regulates Type I IFN-Induced Tolerogenic Signals to Mucosal CD4 T Cells That Attenuate Their Colitogenic Potential
Scientists aimed to investigate the role of CD69 in mucosal immune responses. The expression of CD69 by CD4 T cells isolated from the spleen, mesenteric lymph nodes, small intestinal lamina propria, and colonic lamina propria was determined in specific pathogen-free B6 and TCR transgenic animals, as well as in germ-free B6 mice. [J Immunol] Abstract
