Goblet Cells Deliver Luminal Antigen to CD103+ Dendritic Cells in the Small Intestine
Using a minimally disruptive in vivo imaging approach researchers show that in the steady state, small intestine goblet cells function as passages delivering low molecular weight soluble antigens from the intestinal lumen to underlying CD103+ lamina propria-dendritic cells. Molecular Mechanisms that Control the Expression and Activity of Bcl-6 in TH1 Cells to Regulate Flexibility with a Follicular Helper T Cell-Like Gene Profile
Here researchers found that high concentrations of interleukin 2 inhibited Bcl-6 expression in polarized T helper type 1 cells (TH1 cells). [Nat Immunol] Abstract

Rank Signaling Links the Development of Invariant γδ T Cell Progenitors and Aire+ Medullary Epithelium
Researchers’ data attributed a functional importance to the temporal development of Vγ5+ γδ T cells during thymus medulla formation for αβ T cell tolerance induction and demonstrated a Rank-mediated reciprocal link between dendritic epidermal T cells and autoimmune regulator positive (Aire+) medullary thymic epithelial cell maturation. [Immunity] Abstract

Lung Natural Helper Cells Are a Critical Source of Th2 Cell-Type Cytokines in Protease Allergen-Induced Airway Inflammation
Here investigators report that innate lymphocytes termed lung natural helper cells are a T cell-independent source of T helper 2 (Th2) cell-type cytokines in protease allergen-treated lungs. [Immunity] Abstract

E Protein Transcription Factors Are Required for the Development of CD4+ Lineage T Cells
Here, researchers have demonstrated that in addition to regulating the onset of the double-positive to single-positive transition during T cell development, HEB and E2A also play a separate role in CD4+ lineage choice. [Immunity] Abstract

Transcription Factor Foxo1 Represses T-bet-Mediated Effector Functions and Promotes Memory CD8+ T Cell Differentiation
Herein, scientists demonstrate that inactivation of Foxo1 was essential for instructing T-bet transcription factor-mediated effector differentiation of CD8+ T cells. [Immunity] Abstract

Immune Insufficiency during GvHD Is Due to Defective Antigen Presentation within Dendritic Cell Subsets
Immune suppression due to alloreactivity after transplantation is most profound during graft-versus-host disease (GvHD) after bone marrow transplantation where an inflammatory cytokine storm dominates. Contrary to current dogma, which avers that this is a T-cell defect, researchers demonstrate that the impairment lies within conventional dendritic cells. [Blood] Abstract

Cancer-Associated Epithelial Cell Adhesion Molecule (EpCAM; CD326) Enables Epidermal Langerhans Cell Motility and Migration In Vivo
To gain insight into EpCAM function in a physiologic context in vivo, researchers generated conditional knockout mice with EpCAM-deficient Langerhans cells and characterized them. [Proc Natl Acad Sci USA] Abstract

Signal Transducer and Activator of Transcription-3/Suppressor of Cytokine Signaling-3 (STAT3/SOCS3) Axis in Myeloid Cells Regulates Neuroinflammation
To determine the role of SOCS3 in myeloid cells in neuroinflammation, mice with conditional SOCS3 deletion in myeloid cells were tested for experimental autoimmune encephalomyelitis. [Proc Natl Acad Sci USA] Abstract

Adoptive Transfer with In Vitro Expanded Human Regulatory T Cells Protects Against Porcine Islet Xenograft Rejection via Interleukin-10 in Humanized Mice
This study aimed to determine the ability of human regulatory T cells to prevent islet xenograft rejection and the mechanism(s) involved. [Diabetes] Abstract

A Model for Personalized In Vivo Analysis of Human Immune Responsiveness
Here, researchers demonstrated robust immune reconstitution in mice with hematopoietic stem cells aspirated from bone marrow of adults with type 1 diabetes and healthy control volunteers. [Sci Transl Med]
