Live-Attenuated Vaccine sCPD9 Elicits Superior Mucosal and Systemic Immunity to SARS-CoV-2 Variants in Hamsters

Researchers compared immune responses and preclinical efficacy of the mRNA vaccine BNT162b2, the adenovirus-vectored spike vaccine Ad2-spike, and the live-attenuated virus vaccine candidate sCPD9 in Syrian hamsters, using both homogeneous and heterologous vaccination regimens.

Potent and Selective Covalent Inhibition of the Papain-Like Protease from SARS-CoV-2

The most potent compound inhibited the papain-like protease with kinact/KI= 9,600 M−1 s−1, achieved sub-μM EC50 values against three SARS-CoV-2 variants in mammalian cell lines, and did not inhibit a panel of human deubiquitinases at >30 μM concentrations of inhibitor.

Vaccination Provides Superior In Vivo Recall Capacity of SARS-CoV-2 Specific Memory CD8 T Cells

Researchers compared the memory CD8 T cell response against a common SARS-CoV-2 epitope after vaccination, infection, or both.

Antigen-Specific B Cells Direct T Follicular-Like Helper Cells into Lymphoid Follicles to Mediate Mycobacterium tuberculosis Control

A population of IRF4+ T cells coexpressed the transcription factor BCL6 during Mycobacterium tuberculosis (Mtb) (Mtb) infection, and deletion of BCL6 in CD4+ T cells resulted in reduction of TFH-like cells, impaired localization within granuloma-associated lymphoid tissue, and increased Mtb burden.

    Abstract

NK-Like CD8+ γδ T Cells Are Expanded in Persistent Mycobacterium tuberculosis Infection

The authors analyzed peripheral blood γδ T cells from a South African adolescent cohort and showed that a unique CD8+ γδ T cell subset with features of “memory inflation” expanded in chronic Mycobacterium tuberculosis infection.

Immunization against Zika by Entrapping Live Virus in a Subcutaneous Self-Adjuvanting Hydrogel

Researchers showed that a subcutaneous vaccine consisting of live Zika virus electrostatically entrapped in a self-adjuvanting hydrogel recruited immune cells at the injection site and provided mice with effective protection against a lethal viral challenge.

Pulmonary Inflammation Promoted by Type-2 Dendritic Cells Is a Feature of Human and Murine Schistosomiasis

Human pre-patent Schistosoma mansoni infection pulmonary sputum samples revealed a mixed type-1/type-2 inflammatory cytokine profile, whilst a case-control study showed no significant pulmonary cytokine changes in endemic patent infection.

Interferon-Inducible Phospholipids Govern IFITM3-Dependent Endosomal Antiviral Immunity

Investigators uncovered the requirement of host co-factors for endosomal antiviral inhibition through high-throughput proteomics and lipidomics in cellular models of interferon-induced transmembrane proteins restriction.

MAIT Cells Activate Dendritic Cells to Promote TFH Cell Differentiation and Induce Humoral Immunity

Scientists explored a class of adjuvant that harnessed mucosal-associated invariant T (MAIT) cells, and showed evidence that intranasal immunization of MAIT cell agonists co-administered with protein induced protective humoral immunity and immunoglobulin A production.

    Genotype–Phenotype Landscapes for Immune–Pathogen Coevolution

The authors describe several approaches that probe different regions of the high-dimensional sequence space and comment on how combinations of these methods may offer novel insight into immune–pathogen coevolution.

Interplay between Endogenous and Exogenous Human Retroviruses

Scientists review the impact of infection by exogenous retroviruses on the expression of human endogenous retroviruses (HERVs), the effect of HERVs on the pathogenicity of HIV and human T-cell lymphotropic viruses and on the severity of the diseases caused by them, and more.
