[Nat Commun] Full Article DNA Copy Number Changes Define Spatial Patterns of Heterogeneity in Colorectal Cancer Investigators examined DNA sequence and DNA copy-number heterogeneity in colorectal cancer by targeted high-depth sequencing of 100 most frequently altered genes. In 97 samples, with primary tumors and matched metastases from 27 patients, they observed inter-tumor concordance for coding mutations; in contrast, gene copy numbers are highly discordant between primary tumors and metastases as validated by fluorescent in situ hybridization. [Nat Commun] Full Article Centrosome Amplification Is Sufficient to Promote Spontaneous Tumorigenesis in Mammals Scientists tested the consequence of centrosome amplification by creating mice in which centrosome number can be chronically increased in the absence of additional genetic defects. They showed that increasing centrosome number elevated tumor initiation in a mouse model of intestinal neoplasia. [Dev Cell] Abstract | Graphical Abstract The miR-487b-3p/GRM3/TGFβ Signaling Axis Is an Important Regulator of Colon Cancer Tumorigenesis Researchers demonstrated that expression of GRM3, a metabotropic glutamate receptor mainly expressed in mammalian central nervous system, was significantly upregulated in majority of human colonic adenocarcinomas tested and colon cancer cell lines. [Oncogene] Abstract Given that cortactin controls endothelial permeability, scientists hypothesized that cortactin is also important for epithelial barrier regulation. In healthy human colon tissues, cortactin co-localized with ZO-1 at epithelial cell contacts. In inflammatory bowel diseases patients, they observed decreased cortactin levels and loss of co-localization with ZO-1. [Mucosal Immunol] Abstract Ataxia-telangiectasia mutated (ATM) protein kinase is a major guardian of genomic stability, and its well-established function in cancer is tumor suppression. Researchers reported an oncogenic role of ATM. Using two isogenic sets of human colon cancer cell lines that differed only in their ATM status, they demonstrated that ATM deficiency significantly inhibits cancer cell proliferation, migration, and invasion.
