Characterization of an Engineered Live Bacterial Therapeutic for the Treatment of Phenylketonuria in a Human Gut-on-a-Chip

Gut-on-a-chip microfluidics technology presents an opportunity to characterize strain function within a simulated human gastrointestinal tract. The authors showed how in vitro gut-chip models could be used to construct mechanistic models of strain activity and recapitulate the behavior of the engineered strain in a non-human primate model.

Bifidobacterium dentium-Derived Y-Glutamylcysteine Suppresses ER-Mediated Goblet Cell Stress and Reduces TNBS-Driven Colonic Inflammation

    B. dentium cell-free supernatant and γ-glutamylcysteine were taken up by human colonic T84 cells, increased glutathione levels, and reduced ROS generated by the endoplasmic reticulum (ER)-stressors thapsigargin and tunicamycin.

Salmonella enterica Serovar Typhimurium SPI-1 and SPI-2 Shape the Global Transcriptional Landscape in a Human Intestinal Organoid Model System

Scientists used a human intestinal organoid model to define host transcriptional responses to S. Typhimurium infection, also determining host pathways dependent on Salmonella pathogenicity island- 1-(SPI-1) and -2-encoded type 3 secretion systems

Vitamin D3 Suppresses Intestinal Epithelial Stemness via ER Stress Induction in Intestinal Organoids

Intestinal organoids derived from mouse small intestine were treated with vitamin D3, and the effects on intestinal stemness and differentiation were evaluated using real-time PCR and immunofluorescence staining of the distinct lineage markers.

Cannabinoids Orchestrate Cross-Talk between Cancer Cells and Endothelial Cells in Colorectal Cancer

RNA-sequencing datasets in the NCBI GEO repository were first analyzed; upregulation of cannabinoid receptors was observed in both primary and metastatic colorectal cancer tumor tissues.

Molecular Mechanism of miR-34b-5p and RNA Binding Protein HuR Binding to lncRNA OIP5-AS1 in Colon Cancer Cells

Investigators identified the mechanism of long non-coding RNA OIP5-AS1 (lncRNA OIP5-AS1) in colon cancer (CC) development. OIP5-AS1 expression pattern in CC tissues and cells was detected and the relation between OIP5-AS1 level and CC prognosis was analyzed.

Worenine Reverses the Warburg Effect and Inhibits Colon Cancer Cell Growth by Negatively Regulating HIF-1α

The effects of worenine on colorectal cancer cell proliferation, colony formation and cell cycle distribution were measured.

Metformin Treatment Response Is Dependent on Glucose Growth Conditions and Metabolic Phenotype in Colorectal Cancer Cells

Researchers determined how glucose concentration influences cancer cells’ response to metformin. They used two colorectal cancer cell lines with different growth rates and clinically achievable metformin concentrations.
