TOP STORYScientists demonstrated that therapeutic activation of estrogen receptor beta (ERβ) elicited potent anticancer effects in triple-negative breast cancer through the induction of a family of secreted proteins known as the cystatins, which functioned to inhibit canonical TGFβ signaling and suppress metastatic phenotypes both in vitro and in vivo. PUBLICATIONS(Ranked by impact factor of the journal)The binding of adenosine to adenosine receptor 2B promoted breast cancer stem cell (BCSC) enrichment by activating protein kinase C-δ, which phosphorylated and activated the transcription factor STAT3, leading to increased expression of interleukin 6 and NANOG, two key mediators of the BCSC phenotype. [Proc Natl Acad Sci USA] Abstract Epithelial cell-specific targeting of Mmp14/MT1-MMP in the normal mammary gland failed to impair branching, whereas deleting the proteinase in carcinoma cells abrogated invasion, preserved matrix architecture, and completely blocked metastasis. [Dev Cell] Full Article | Graphical Abstract Overexpression of semaphorin 7a (SEMA7A) promoted the presence of myeloid-derived podoplanin (PDPN)-expressing cells in the tumor microenvironment and during involution. Mammary tissue from SEMA7A-knockout mice exhibited decreased myeloid-derived PDPN-expressing cells, PDPN-expressing endothelial cells, and lymphatic vessel density. [Cancer Res] Abstract C/EBPδ Links IL-6 and HIF-1 Signaling to Promote Breast Cancer Stem Cell-Associated Phenotypes Investigators showed that deletion or depletion of C/EBPδ reduced expression of stem cell factors and stemness markers, sphere formation and self-renewal, along with growth of tumors and established experimental metastases in vivo. [Oncogene] Abstract In vitro, trifluoperazine (TFP) induced G0/G1 cell cycle arrest via decreasing the expression of both cyclinD1/CDK4 and cyclinE/CDK2, and stimulated mitochondria-mediated apoptosis. In vivo, TFP suppressed the growth of subcutaneous xenograft tumor and brain metastasis without causing detectable side effects. [Cell Death Dis] Full Article PYK2 Negatively Regulates the Hippo Pathway in TNBC by Stabilizing TAZ Protein Researchers found that inhibition of PYK2 expression or its kinase activity substantially affected the steady-state level of transcriptional coactivator with PDZ-binding motif (TAZ) and markedly facilitated its proteasomal degradation. [Cell Death Dis] Full Article Overexpression of TFPI-2 decreased phosphorylation of ERK1/2 and the translocation of p-ERK1/2 from cytoplasm into the nucleus, and eventually resulted in reduced cell proliferation. The authors identified myosin-9 and actinin-4 as TFPI-2-interacting proteins. Full-length TFPI-2 was required for binding to actinin-4, whereas the N + KD1 regions of TFPI-2 were sufficient to interact with myosin-9. [Sci Rep] Full Article Scientists report that proteasome-associated deubiquitinase inhibitors b-AP15 and platinum pyrithione induced growth inhibition in estrogen receptor α (ERα) positive breast cancer cells. They showed that these inhibitors induced cell cycle arrest and apoptosis associated with caspase activation, endoplasmic reticulum stress and the downregulation of ERα. [Oncogenesis] Full Article The response of 3D-derived MCF-7 cells subsequently cultured in 2D in medium supplemented with glutathione to Doxorubicin (DOX) was evaluated. These cells demonstrated a resistance towards DOX closer to that displayed by 3D spheroids, which was higher than that exhibited by standard 2D cell cultures.
