TOP STORYRET Rearrangements Are Actionable Alterations in Breast Cancer Researchers reported RET gene alterations, including amplification, missense mutations, known fusions, novel fusions, and rearrangements in breast cancer. Their frequency, oncogenic potential, and actionability in breast cancer were described. [Nat Commun] Full Article

PUBLICATIONS(Ranked by impact factor of the journal)Scientists performed single-cell transcriptome analysis on ERα-positive BCa cells following 17β-estradiol stimulation and reconstructed the dynamic estrogen-responsive transcriptional network from discrete time points into a pseudotemporal continuum. [Cell Rep] Full Article | Graphical Abstract Janus Kinase 1 Plays a Critical Role in Mammary Cancer Progression Investigators showed that janus kinase 1 was essential for IL-6-class inflammatory cytokine signaling and played a critical role in metastatic cancer progression. [Cell Rep] Full Article | Graphical Abstract Oncogenic Activity of Poly (ADP-Ribose) Glycohydrolase Using both in vitro and in vivo models, researchers demonstrated that heightened expression of catalytically active poly (ADP-ribose) glycohydrolase facilitated cell transformation and invasion of normal mammary epithelial cells. [Oncogene] Full Article In independent functional expression studies in triple-negative breast cancer models, ephrin type-A receptor 2 (EPHA2) gene-silencing combined with eicosapentaenoic acid significantly reduced cell growth and enhanced apoptosis compared with monotherapies, both in vitro and in vivo. [Oncogene] Abstract Researchers demonstrated that apigenin significantly suppressed the proliferation and migration of triple-negative breast cancer cells. [Cell Death Discov] Full Article NMT1 Inhibition Modulates Breast Cancer Progression through Stress-Triggered JNK Pathway Scientists demonstrated that genetic inhibition of N-myristoyltransferase NMT1 suppressed initiation, proliferation and invasion of breast cancer cells either in vitro or in vivo. [Cell Death Dis] Full Article Forced expression of trefoil factor 3 in immortalized-human mammary epithelial cells enhanced cell proliferation, cell survival, anchorage-independent growth, produced highly disorganized three-dimensional acinar structures and generated tumors in immunocompromized mice. [Cell Death Dis] Full Article Effects of Modulating Actin Dynamics on HER2 Cancer Cell Motility and Metastasis The growth and viability of human HER2+ breast cancer and ovarian cancer cell lines were significantly impaired upon treatment with the marine macrolide mycalolide B. [Sci Rep] Full Article Induction of N-Ras Degradation by Flunarizine-Mediated Autophagy To target N-Ras in basal-like breast cancer, investigators screened existing pharmacologically active compounds for the new ability to induce N-Ras degradation, which led to the identification of flunarizine, previously approved for treating migraine and epilepsy. [Sci Rep] Full Article Water-pipe smoking (WPS) induced epithelial-mesenchymal transition of MCF7 and BT20 breast cancer cell lines; thus, WPS enhanced cell invasion ability of both cell lines in comparison with their matched controls. REVIEWSCrosstalk between Estrogen Signaling and Breast Cancer Metabolism The authors discuss studies revealing the role of estrogen signaling elements in drug resistance development and metabolic adaptation during breast cancer progression. [Trends Endocrinol Metab] Abstract Calcium Signaling and Breast Cancer Investigators consider calcium signaling in the context of breast cancer and provide an overview of the roles that have been attributed to specific regulators of cellular calcium levels in processes relevant to breast cancer progression. INDUSTRY NEWSHusband-and-Wife Doctors Receive $1.8 Million to Test New Breast Cancer Approach Radiation oncologist Timothy Showalter, MD, and breast cancer surgeon Shayna L. Showalter, MD, are leading an interdisciplinary effort to evaluate a technique they have developed at the University of Virginia called Precision Breast Intraoperative Radiation Therapy. [University of Virginia] Press Release Two New Grants to Fuel Metastatic Breast Cancer Research Dr. Erica Goddard, a postdoctoral research fellow in Dr. Cyrus Ghajar’s Laboratory for the Study of Metastatic Microenvironments, won a Breakthrough Fellowship Award from the Department of Defense Breast Cancer Research Program. She will receive a total of $528,000 over three years. [Fred Hutchinson Cancer Research Center] Press Release

POLICY NEWSAI Peer Reviewers Unleashed to Ease Publishing Grind A suite of automated tools is now available to assist with peer review but humans are still in the driver’s seat. A handful of academic publishers are piloting AI tools to do anything from selecting reviewers to checking statistics and summarizing a paper’s findings. [Nature News] Editorial Stop Exploitation of Foreign Postdocs in the United States A survey reveals some lab heads are using the need for visas to create unacceptable conditions for junior researchers. Some PIs are exploiting the fact that overseas scientists rely on them for continued visas. The responses suggest that senior scientists are using this reliance to force postdocs to work longer hours and endure unacceptable conditions. [Nature] Editorial
