Avasopasem Manganese Synergizes with Hypofractionated Radiation to Ablate Tumors through the Generation of Hydrogen Peroxide

In vitro non-small lung cancer and mammary adenocarcinoma models showed that auranofin-induced inhibition of glutathione- and thioredoxin-dependent hydrogen peroxide metabolism selectively enhanced Avasopasem manganese-induced killing of cancer cells compared to normal cells.

FUT8-Mediated Aberrant N-Glycosylation of B7H3 Suppresses the Immune Response in Triple-Negative Breast Cancer

Researchers dissected the mechanisms by which TNBC cells initiate immunosuppression by inducing glycosylation B7 homolog 3 protein (B7H3) stabilization, and suggested the fucosyltransferase (FUT8)-B7H3 axis as a potential prognostic biomarker and therapeutic target for TNBC patients.

Disruption of FOXO3a-miRNA Feedback Inhibition of IGF2/IGF-1R/IRS1 Signaling Confers Herceptin Resistance in HER2-Positive Breast Cancer

The authors demonstrated that the insulin-like growth factor/ insulin-like growth factor-1/ receptor/ insulin receptor substrate-1 (IGF2/IGF-1R/IRS1) signaling was aberrantly activated in Herceptin-resistant breast cancer via disruption of the FOXO3a-miRNA negative feedback inhibition.

Bedaquiline, an FDA-Approved Drug, Inhibits Mitochondrial ATP Production and Metastasis In Vivo, by Targeting the Gamma Subunit (ATP5F1C) of the ATP Synthase

Scientists found that Bedaquiline had no effect on the growth of non-tumorigenic mammary epithelial cells or primary tumors in vivo.

Cryopreservation of Human Cancers Conserves Tumor Heterogeneity for Single-Cell Multi-Omics Analysis

Using the Chromium 10X platform, investigators sequenced a total of ~ 120,000 cells from fresh and cryopreserved replicates across three primary breast cancers, two primary prostate cancers and a cutaneous melanoma.

PI5P4Ks Drive Metabolic Homeostasis through Peroxisome-Mitochondria Interplay

Researchers identified a role for peroxisomal phosphatidylinositol 4,5-bisphosphate generated by the phosphatidylinositol-5-phosphate 4-kinases (PI5P4Ks) in maintaining energy balance, and noted that inhibition of the PI5P4Ks in cancer cells led to decreased cell viability.

Targeting Epigenetic and Post-Transcriptional Gene Regulation by PSF Impairs Hormone Therapy-Refractory Cancer Growth

Investigators conducted an in vitro chemical array screen and identified multiple small molecules that interacted with PSF. Several molecules inhibited RNA binding by PSF and decreased prostate cancer cell viability.

The N Terminus of Orai1 Couples to the AKAP79 Signaling Complex to Drive NFAT1 Activation by Local Ca2+ Entry

Researchers assessed the coupling of Orai1 Ca2+ channels with nuclear factor of activated T cells in MCF-7 cells to help identify an approach for generation of immunosuppressant drugs.

PI(3,4)P2-Mediated Membrane Tubulation Promotes Integrin Trafficking and Invasive Cell Migration

Scientists used the invadopodium model system of Src-transformed fibroblast and MDA-MB-231 cell lines to investigate the functional role of PI(3,4)P2 in adhesion signaling.

The Extracellular-Regulated Protein Kinase 5 (ERK5) Enhances Metastatic Burden in Triple-Negative Breast Cancer through Focal Adhesion Protein Kinase (FAK)-Mediated Regulation of Cell Adhesion

The authors revealed that ERK5 was critical for maintaining the invasive capability of TNBC cells through FAK activation, and reported that a high level of ERK5 is a predictive marker of metastatic breast cancer.

Novel miR-5088-5p Promotes Malignancy of Breast Cancer by Inhibiting DBC2

The relationship between Deleted in Breast Cancer 2 (DBC2) and miR-5088-5p was analyzed by scientists, and both of which were suggested as potential novel biomarkers for the diagnosis of breast cancer patients.

Myeloid-Derived Suppressor Cells Regulate the Immunosuppressive Functions of PD-1−PD-L1+ Bregs through PD-L1/PI3K/AKT/NF-κB Axis in Breast Cancer

Researchers reported that blocking PD-1/PD-L1 interaction between myeloid-derived suppressor cells and B cells could reverse the immunosuppressive functions of PD-1−PD-L1+ Bregs.

CXCR4 Intracellular Protein Promotes Drug Resistance and Tumorigenic Potential by Inversely Regulating the Expression of Death Receptor 5

Scientists revealed that the presence of CXCR4 intracellular protein augments paclitaxel resistance and pro-tumorigenic functions using breast and colon cancer cells lines, and that targeting this protein may be critical to dampen the pro-tumorigenic functions of CXCR4.
