DIF-1 Inhibits Growth and Metastasis of Triple-Negative Breast Cancer through AMPK-Mediated Inhibition of the mTORC1-S6K Signaling Pathway

Scientists found that differentiation-inducing factor-1 (DIF-1) induced phosphorylation of AMPK and Raptor and dephosphorylation of p70S6K in multiple TNBC cell lines.

MICAL1 Regulates Actin Cytoskeleton Organization, Directional Cell Migration and the Growth of Human Breast Cancer Cells as Orthotopic Xenograft Tumours

MICAL1 gene disruption by CRISPR/Cas9 in MDA MB 231 human breast cancer cells knocked out protein expression, which affected F-actin organization, cell size and motility.

ETV7 Regulates Breast Cancer Stem-Like Cell Features by Repressing IFN-Response Genes

Researchers showed a new role for ETV7, a transcriptional repressor member of the ETS family, in promoting breast cancer stem-like cells plasticity and resistance to chemo- and radiotherapy in breast cancer cells.

C5aR1-Positive Neutrophils Promote Breast Cancer Glycolysis through WTAP-Dependent m6A Methylation of ENO1

C5aR1-positive neutrophil-secreted IL1β and TNFα cooperatively activated  ERK1/2 signaling, which phosphorylated WTAP at serine341 and thereby stabilized WTAP protein. The stabilization of WTAP further promoted  RNA m6A methylation of ENO1, impacting the glycolytic activity of breast cancer cells.

Sublethal Doxorubicin Promotes Migration and Invasion of Breast Cancer Cells: Role of Src Family Non-Receptor Tyrosine Kinases

Investigators  showed that sublethal doxorubicin treatment led to increased migration and invasion in otherwise non-invasive MCF7 breast cancer cells.

Mutant p53 Attenuates Oxidative Phosphorylation and Facilitates Cancer Stemness through Downregulating microRNA-200c-PCK2 Axis in Basal-Like Breast Cancer

The authors demonstrated that CRISPR-mediated knockout of miR-200c induced metabolic features similar to the metabolic rewiring caused by p53 hot-spot mutations, and that impairing this metabolic reprogramming interfered with miR-200c deficiency-induced stemness and transformation.

A New Immunochemical Strategy for Triple-Negative Breast Cancer Therapy

Following conjugation to β-lactam-derivatized monomethyl auristatin F, tropomyosin receptor kinase B cell surface protein-targeting dual variable domain-antibody–drug conjugates showed potency against multiple breast cancer cell lines, including TNBC cell lines.

Context-Dependent Effects of Whole-Genome Duplication during Mammary Tumor Recurrence

In recurrent tumor cells, whole-genome duplication (WGD) slowed tumor formation, growth rate and opposed the process of recurrence, while WGD promoted the growth of primary tumors.

Knockdown of the Prognostic Cancer Stem Cell Marker Musashi-1 Decreases Radio-Resistance While Enhancing Apoptosis in Hormone Receptor-Positive Breast Cancer Cells via p21WAF1/CIP1

Musashi-1 was artificially downregulated in MCF-7 breast cancer cells and implications for cancer stem cell markers, cell apoptosis and apoptosis regulator p21, proliferation and radiation response were analyzed via flow cytometry and colony formation.

Molecular Imaging Reveals a High Degree of Cross-Seeding of Spontaneous Metastases in a Novel Mouse Model of Synchronous Bilateral Breast Cancer

Researchers engineered human breast cancer cells to express either Akaluc or Antares2 for bioluminescence imaging and tdTomato or zsGreen for ex vivo fluorescence microscopy.

Single-Cell Immunoblotting Resolves Estrogen Receptor-α Isoforms in Breast Cancer

To scrutinize hormone response heterogeneity among breast cancer (BCa) tumor cells, scientists developed a precision tool to specifically measure estrogen receptor alpha (ER-α)-66, ER-α46, and eight ER-signaling proteins with single-cell resolution in the highly hetero-clonal MCF-7 BCa cell line.

Arginine Depriving Enzymes: Applications as Emerging Therapeutics in Cancer Treatment

The L-Arginine (Arg) auxotrophic cancerous cells like hepatocellular carcinoma, human colon cancer, leukemia, and breast cancer cells were sensitive to Arg depriving enzymes treatment due to low expression of crucial enzymes argininosuccinate synthetase, argininosuccinate lyase, and ornithine transcarbamylase.
