N-acetylcysteine Overcomes NF1 Loss-Driven Resistance to PI3Kα Inhibition in Breast Cancer

Depletion of NF1 in PIK3CAH1047R breast cancer cell lines and a patient-derived organoid model showed that NF1 loss reduced sensitivity to phosphatidylinositol 3-kinase α (PI3Kα) inhibition and correlated with enhanced glycolysis and lower levels of reactive oxygen species.

ER-Localized JmjC Domain-Containing Protein JMJD8 Targets STING to Promote Immune Evasion and Tumor Growth in Breast Cancer

Researchers demonstrated that an endoplasmic reticulum (ER)-localized JmjC domain-containing protein, JMJD8, inhibited STING-induced type I interferon responses to promote immune evasion and breast tumorigenesis.

eIF2Bδ Blocks the Integrated Stress Response and Maintains eIF2B Activity and Cancer Metastasis by Overexpression in Breast Cancer Stem Cells

The authors described a unique translation regulatory axis enacted by reduced expression of miR-183 in breast cancer stem cells, which they showed targeted the eIF2Bδ subunit of guanine nucleotide exchange factor eIF2B.

    Abstract

ETV7 Reduces Inflammatory Responses in Breast Cancer Cells by Repressing the TNFR1/NF-κB Axis

Scientists identified TNFRSF1A, encoding for the main receptor of TNF-α, TNFR1, as one of the genes down-regulated by ETV7. They showed that ETV7 directly bound to the intron I of this gene, and that the ETV7-mediated down-regulation of TNFRSF1A reduced the activation of NF-κB signaling.

RON-Augmented Cholesterol Biosynthesis in Breast Cancer Metastatic Progression and Recurrence

Investigators modeled breast cancer recurrence using implantation of RON-overexpressing murine breast cancer cells. Recurrent growth was examined after tumor resection via in vivo imaging and ex vivo culture of circulating tumor cells from whole blood samples from tumor bearing mice.

Suppression of Triple-Negative Breast Cancer Aggressiveness by LGALS3BP via Inhibition of the TNF-α–TAK1–MMP9 Axis

The authors investigated the function and underlying mechanism of action of lectin galactoside-binding soluble 3 binding protein (LGALS3BP) in TNBC progression and determined the therapeutic potential of nanoparticle-mediated delivery of LGALS3BP in TNBC.

PDK1-Stabilized LncRNA SPRY4-IT1 Promotes Breast Cancer Progression via Activating NF-κB Signaling Pathway

Researchers found that lncRNA sprouty4-intron transcript 1 (SPRY4-IT1) was regulated by pyruvate dehydrogenase kinase 1 (PDK1) with correlation analysis, and PDK1 upregulated SPRY4-IT1 remarkably in breast cancer cells.

    Abstract

Circular Forms of Dedicator of Cytokinesis 1 Promotes Breast Cancer Progression by Derepressing Never in Mitosis Related Kinase 2 via Sponging miR-128-3p

Scientists explored the regulation between circular forms of dedicator of cytokinesis 1 and miR-128-3p, and its implication on the pathogenesis of breast cancer modulated by never in mitosis related kinase 2.

    Abstract

The Estrogen/miR-338-3p/ADAM17 Axis Enhances the Viability of Breast Cancer Cells via Suppressing NK Cell’s Function

miR-338-3p level was found to be significantly downregulated in breast cancer (BC) tissues and estrogen receptor positive (ER+) cells, this difference was more obvious in ER+ patients or BC patients at advanced stage.

Triumphs and Challenges in Exploiting poly(ADP-Ribose) Polymerase Inhibition to Combat Triple-Negative Breast Cancer

The authors discuss the roles of poly(ADP-ribose) polymerase (PARP) in mediating DNA repair processes in breast epithelial cells, mechanisms of PARPi resistance in TNBC, and recent advances in the development of agents designed to overcome PARPi resistance in TNBC.
