    TOP STORY

Tests For Genes Don’t Predict Breast Cancer Better
Studying genes linked to breast cancer may someday lead to better treatments, but they do little to improve a doctor’s ability to predict who is likely to develop a tumor, researchers reported. [Press release from Reuters discussing online prepublication in The New England Journal of Medicine]

    SCIENCE NEWS

Another Perk of Painkillers? Decreased Hormone Levels May Reduce Cancer Risk
Postmenopausal women who regularly use aspirin and other analgesics have lower estrogen levels, which could contribute to a decreased risk of breast or ovarian cancer. [Press release from ScienceDaily discussing online prepublication in Cancer Epidemiology, Biomarkers & Prevention]

The Biomarkers Consortium Launches I-SPY 2 Breast Cancer Clinical Trial
The Biomarkers Consortium announced the launch of a highly anticipated clinical trial to help screen promising new drugs being developed for women with high risk, fast-growing breast cancers – women for whom an improvement over standard treatment could dramatically change the odds of survival. [The Biomarkers Consortium Press Release]

Scott & White Healthcare Researchers Studying Investigational Agent that Targets Breast Cancer
Researchers are studying an investigational agent that targets metastatic breast cancer to find if it can stop cancer cells from continuing to grow. [EurekAlert! Press Release]

    CURRENT PUBLICATIONS

    LABORATORY RESEARCH

Dose-Dependent Induction of Distinct Phenotypic Responses to Notch Pathway Activation in Mammary Epithelial Cells
Researchers investigated the cellular basis for the heterogeneous responses to Notch pathway activation in 3D cultures of MCF-10A mammary epithelial cells. [Proc Natl Acad Sci U S A]

Integrin-Linked Kinase has a Critical Role in ErbB2 Mammary Tumor Progression: Implications for Human Breast Cancer
Here, researchers show that mammary epithelial disruption of integrin-linked kinase results in a profound block in mammary tumor induction. [Oncogene]

Artemin is Estrogen Regulated and Mediates Antiestrogen Resistance in Mammary Carcinoma
Researchers report that artemin (ARTN) is an estrogen-inducible gene. Meta-analysis of gene expression data sets showed that ARTN expression is positively correlated to estrogen receptor status in human mammary carcinoma. [Oncogene]

Potential Prognostic Value of Heat-Shock Protein 90 in the Presence of Phosphatidylinositol-3-Kinase Overexpression or Loss of PTEN, in Invasive Breast Cancers
Evaluating the expression of signaling molecule proteins from the mitogen-activated protein kinase pathway and the phosphatidylinositol-3-kinase pathway in invasive breast cancers may identify prognostic marker(s) associated with early relapse. [Breast Cancer Res]

Ack1 Mediated AKT/PKB Tyrosine 176 Phosphorylation Regulates Its Activation
Researchers have uncovered that growth factors binding to receptor tyrosine kinases lead to activation of a non-receptor tyrosine kinase, Ack1 (also known as ACK or TNK2), which directly phosphorylates AKT at an evolutionarily conserved tyrosine 176 in the kinase domain. [PLoS ONE]

Transient Tumor-Fibroblast Interactions Increase Tumor Cell Malignancy by a TGF-Beta Mediated Mechanism in a Mouse Xenograft Model of Breast Cancer
Here, researchers report a role for normal stroma fibroblasts in the progression of invasive tumors to metastatic tumors. [PLoS ONE]

Common Germ-Line Polymorphism of C1QA and Breast Cancer Survival
The purpose of this study was to investigate the association of single nucleotide polymorphism, rs172378, with expression of C1QA and breast cancer survival. [Br J Cancer]

    CLINICAL RESEARCH

Performance of Common Genetic Variants in Breast-Cancer Risk Models
The inclusion of newly discovered genetic factors modestly improved the performance of risk models for breast cancer. The level of predicted breast-cancer risk among most women changed little after the addition of currently available genetic information. [N Engl J Med]

Breast Cancer Among the Oldest Old: Tumor Characteristics, Treatment Choices, and Survival
Women 80 years and older have breast cancer characteristics similar to those of younger women yet receive less aggressive treatment and experience higher mortality from early-stage breast cancer.  [J Clin Oncol]

Radiation Exposure, the ATM Gene, and Contralateral Breast Cancer in the Women’s Environmental Cancer and Radiation Epidemiology Study
Women who carry rare deleterious ATM missense variants and who are treated with radiation may have an elevated risk of developing contralateral breast cancer. [J Natl Cancer Inst]

Analgesic Use and Sex Steroid Hormone Concentrations in Postmenopausal Women
Prior epidemiologic studies suggest that regular use of analgesics may decrease risk of breast and ovarian cancer.  Researchers explored possible hormone-mediated mechanisms for these associations by examining the relationship between use of aspirin, nonaspirin nonsteroidal anti-inflammatory drugs, and acetaminophen and sex steroid hormone concentrations among 740 postmenopausal women in the Nurses’ Health Study.
