SCIENCE NEWS Endocrine Sensitivity Measure Predicts Breast Cancer Survival A patient’s sensitivity to endocrine therapy index, a measure of estrogen receptor-related transcription activity, appears to be predictive of distant relapse-free survival in breast cancer patients who receive endocrine therapy alone or endocrine therapy after chemotherapy, but not in those who receive no adjuvant therapy. [Press release from ModernMedicine discussing online prepublication in the Journal of Clinical Oncology] No Benefit from Infusional Chemotherapy in Early Breast Cancer For patients with early-stage breast cancer, adjuvant treatment with intravenous epirubicin, cisplatin, and 5-fluorouracil shows no benefit in terms of reducing relapse rates over 5-fluorouracil, epirubicin, and cyclophosphamide. [Press release from MedWire News discussing online prepublication in Annals of Oncology] Breast Cancer Risk Varies Among Different Progestins Used in Hormone Replacement Therapy Researchers compared four types of progestins used in hormone replacement therapies and found significantly different outcomes on the progression of breast cancer in an animal model depending on the type of progestin used. [Press release from EurekAlert! discussing online prepublication in Cancer Prevention Research] Postmenopausal Therapy Poses Risk for Breast Cancer Using postmenopausal hormone therapy for more than 15 years increases a woman’s risk of developing breast cancer, particularly among leaner women. [Press release from the University of Southern California discussing online prepublication in Cancer Epidemiology, Biomarkers and Prevention] K-State Research Team Investigates Mutated Gene’s Role in Breast Cancer A microscopic gene may play a gigantic role when it comes to cancerous tissue in the human body. The team is investigating mutations within the ADAM12 gene of the A Disintegrin and Metalloprotease family, or ADAM family, and its role in breast cancer. [Kansas State University Press Release]

CURRENT PUBLICATIONS (Ranked by Impact Factor of the Journal) LABORATORY RESEARCH The Protein Kinase Pak4 Disrupts Mammary Acinar Architecture and Promotes Mammary Tumorigenesis In this study investigators used immortalized mouse mammary epithelial cells as a model system to study the role for Pak4 in mammary tumorigenesis. [Oncogene] Evi dence for a Pro-Apoptotic Function of RACK1 in Human Breast Cancer Scientists have recently conducted an analysis of RACK1 messenger RNA expression in 127 human breast cancer and 33 non-cancerous breast tissue specimens. [Oncogene] Synthetic Progestins Differentially Promote or Prevent 7,12-Dimethylbenz(a)anthracene-Induced Mammary Tumors in Sprague-Dawley Rats The goal of this study was to compare the effects of four clinically relevant progestins, medroxyprogesterone acetate, norgestrel, norethindrone, and megestrol acetate, on 7,12-dimethylbenz(a)anthracene-induced mammary carcinogenesis in the rat. [Cancer Prev Res] Genetic Manipulation of Individual Somatic Mammary Cells In Vivo Reveals a Master Role of S TAT5a in Inducing Alveolar Fate Commitment and Lactogenesis in the Absence of Reproductive Hormones By exploiting an intraductal retrovirus-mediated gene delivery method to introduce a marker gene, researchers found that ductal epithelial cells are turned over with a half time of approximately one month in adult virgin mice. [Dev Biol] Redefining the Expression and Function of the Inhibitor of Differentiation 1 in Mammary Gland Development Generation and analysis of a transgenic mouse model of Inhibitor of Differentiation 1 (Id1) overexpression in the mammary gland reveals that Id1 is insufficient for neoplastic progression in virgin animals or to prevent terminal differentiation of the luminal epithelia during pregnancy and lactation. Together, these data demonstrated that there is no luminal cell-autonomous role for Id1 in mammary epithelial cell fate determination, ductal morphogenesis and terminal differentiation. [PLoS One] Transforming Growth Factor-Beta 1-Mediated Activation of NF-Kappa B Contributes to Enhanced ADAM-12 Expression in Mammary Carcinoma Cells The study identified a cellular mechanism for induction of A disintegrin and metalloproteinase-12 (ADAM-12), which involves nuclear factor-kappa B (NF-kappa B) and its activation by transforming growth factor-beta 1 (TGF-beta1). [Mol Cancer Res] CLINICAL RESEARCH Genomic Index of Sensitivity to Endocrine Therapy for Breast Cancer Scientists hypothesized that measurement of gene expression related to estrogen receptor-alpha (ESR1) within a breast cancer sample represents intrinsic tumoral sensitivity to adjuvant endocrine therapy. [J Clin Oncol] A Randomised Comparative Trial of Infusional ECisF Versus Conventional FEC as Adjuvant Chemotherapy in Early Breast Cancer: The TRAFIC Trial In this multicentre, open-label, phase III randomised controlled trial, 349 women were randomly assigned to receive i.v. ECisF [epirubicin 60 mg/m(2), day 1, cisplatin 60 mg/m(2), day 1 and 5-FU 200 mg/m(2) by daily 24-h infusion (n = 172)] or FEC [5-FU 600 mg/m(2), day 1, epirubicin 60 mg/m(2), day 1 and cyclophosphamide 600 mg/m(2), day 1 (n = 177)]. [Ann Oncol] Menopausal Hormone Therapy and Subsequent Risk of Specific Invasive Breast Cancer Subtypes in the California Teachers Study Researchers investigated hormone therapy use in relation to breast cancer risk in the California Teachers Study cohort.
