    TOP STORY

Repression of Mammary Stem/Progenitor Cells by P53 is Mediated by Notch and Separable from Apoptotic Activity  ShareThis
The results demonstrate that basal levels of p53 restrict mammary stem/progenitor cells through Notch and that the Notch pathway is a therapeutic target to prevent expansion of this vulnerable pool of cells. [Stem Cells]

    SCIENCE NEWS

Nanoshells Provide Golden ‘Touch’ in Killing Breast Tumors
Using tiny gold “nanoshells” to deliver just a little heat to breast tumor cells already treated with radiation boosts the killing potential of the treatment – not just shrinking the tumor but killing the cancer stem cells, said researchers. [Press release from the Baylor College of Medicine discussing online prepublication in Science Translational Medicine]

Nanodrug Enables Image-Guided Breast Cancer Therapy
By combining an iron oxide nanoparticle, a tumor-targeting peptide, and a therapeutic nucleic acid into one construct, a team of investigators has created an agent that holds potential as targeted therapy for breast cancer. [Press release from the National Cancer Institute discussing online prepublication in Cancer Research]

Fox Chase Researchers Identify Risk Factors for the Spread of Breast Cancer to Lymph Nodes
By carefully examining recurrence patterns of thousands of women with breast cancer from records spanning more than 30 years, researchers have now shown that the appearance of lymphovascular invasion in breast tissue predicts the future recurrence of cancer to nearby lymph nodes. [Press release from the Fox Chase Cancer Center discussing data presented at the Annual Meeting of the American Society for Radiation Oncology]

CURRENT PUBLICATIONS (Ranked by Impact Factor of the Journal)

    LABORATORY RESEARCH

Magnetically Vectored Nanocapsules for Tumor Penetration and Remotely Switchable On-Demand Drug Release
Nanocapsules containing intentionally trapped magnetic nanoparticles and defined anticancer drugs have been prepared to provide a powerful magnetic vector under moderate gradient magnetic fields. [Nano Lett]

Autocrine TGF-Beta and Stromal Cell-Derived Factor-1 (SDF-1) Signaling Drives the Evolution of Tumor-Promoting Mammary Stromal Myofibroblasts
Using a coimplantation breast tumor xenograft model, researchers show that resident human mammary fibroblasts progressively convert into carcinoma-associated fibroblasts myofibroblasts during the course of tumor progression. [Proc Natl Acad Sci USA]

The Mitotic Kinase Aurora-A Induces Mammary Cell Migration and Breast Cancer Metastasis by Activating the Cofilin-F-actin Pathway
Here, researchers report that the mitotic kinase Aurora-A (Aur-A) promotes breast cancer metastasis. Aur-A overexpression enhanced mammary cell migration by dephosphorylation and activation of cofilin, which facilitates actin reorganization and polymerization. [Cancer Res]

Image-Guided Breast Tumor Therapy Using a Small Interfering RNA Nanodrug
In this study, researchers describe the synthesis and testing of a tumor-targeted nanodrug (MN-EPPT-siBIRC5) that is designed to specifically shuttle siRNA to human breast tumors. [Cancer Res]

Endoglin Expression in Breast Tumor Cells Suppresses Invasion and Metastasis and Correlates with Improved Clinical Outcome
Here, researchers demonstrate that endoglin is expressed in a subset of invasive breast cancers and cell lines and is subject to epigenetic silencing by gene methylation. [Oncogene]

P-Rex1 Participates in Neuregulin-ErbB Signal Transduction and its Expression Correlates with Patient Outcome in Breast Cancer
Here, researchers report the identification of the guanine nucleotide exchange factor, phosphatidylinositol 3,4,5-triphosphate-dependent Rac exchanger 1 (P-Rex1), as a novel mediator in signaling by ErbB/HER receptors. [Oncogene]

CdGAP is Required for Transforming Growth Factor Beta- and Neu/ErbB-2-Induced Breast Cancer Cell Motility and Invasion
In this study, researchers found that Cdc42 GTPase-activating protein (CdGAP) and mRNA levels are highly increased in mammary tumor explants expressing an activated Neu/ErbB-2 receptor. [Oncogene]

Thermal Enhancement with Optically Activated Gold Nanoshells Sensitizes Breast Cancer Stem Cells to Radiation Therapy
Findings suggest that local hyperthermia delivered by gold nanoshells plus radiation can eliminate radioresistant breast cancer stem cells. [Sci Transl Med]

    CLINICAL RESEARCH

Independent Association of Lobular Involution and Mammographic Breast Density With Breast Cancer Risk
To evaluate whether lobular involution and mammographic breast density are independently associated with breast cancer risk in women with benign breast disease, researchers performed a nested cohort study among women (n = 2666) with benign breast disease diagnosed at Mayo Clinic between January 1, 1985, and December 31, 1991 and a mammogram available within 6 months of the diagnosis. [J Natl Cancer Inst]

ANCCA/ATAD2 Overexpression Identifies Breast Cancer Patients with Poor Prognosis, Acting to Drive Proliferation and Survival of Triple-Negative Cells through Control of B-Myb and EZH2
In this study, researchers show that ANCCA is overexpressed in >70% of breast tumors and that its high protein level correlates well with tumor histologic grades (P < 0.0001), highlighting ANCCA as a prognostic factor for poor overall survival and disease recurrence. [Cancer Res]
