SCIENCE NEWS News Note: Scientists Identify Molecular Link Between BRCA1 Protein Levels and Obesity NCI researchers have defined a possible molecular link between breast cancer risk and obesity. New study results show that a protein called C-terminal binding protein (CtBP) acts to control a gene linked to breast cancer risk in rapidly growing cells by monitoring and responding to how the cells use and store energy (metabolic state). CURRENT PUBLICATIONS (Ranked by Impact Factor of the Journal) LABORATORY RESEARCH Magnetically Vectored Nanocapsules for Tumor Penetration and Remotely Switchable On-Demand Drug Release Nanocapsules containing intentionally trapped magnetic nanoparticles and defined anticancer drugs have been prepared to provide a powerful magnetic vector under moderate gradient magnetic fields. In vitro as well as in vivo results indicate that these nanocapsules can be enriched near the mouse breast tumor and are effective in reducing tumor cell growth. [Nano Lett] Nucleotide Excision Repair Deficiency Is Intrinsic in Sporadic Stage I Breast Cancer Taken together, the data suggest that nucleotide excision repair (NER) deficiency may play an important role in the etiology of sporadic breast cancer, and that early-stage breast cancer may be intrinsically susceptible to genotoxic chemotherapeutic agents, such as cis-platinum, whose damage is remediated by NER. [Proc Natl Acad Sci USA] Profilin1 Regulates PI(3,4)P2 and Lamellipodin Accumulation at the Leading Edge Thus Influencing Motility of MDA-MB-231 Cells In this study, researchers show that profilin1 negatively regulates lamellipodin targeting to the leading edge in MDA-MB-231 breast cancer cells and normal cells; profilin1 depletion increases lamellipodin concentration at the lamellipodial tip (where it binds Ena/VASP), and this mediates the hypermotility. [Proc Natl Acad Sci USA] Systematic Detection of Putative Tumor Suppressor Genes Through the Combined Use of Exome and Transcriptome Sequencing To identify potential tumor suppressor genes, genome-wide data from exome and transcriptome sequencing were combined to search for genes with loss of heterozygosity and allele specific expression. The analysis was conducted on the breast cancer cell line, HCC1954, and a lymphoblast cell line from the same individual, HCC1954BL. [Genome Biol] Evidence for SMAD3 as a Modifier of Breast Cancer Risk in BRCA2 Mutation Carriers Current attempts to identify genetic modifiers of BRCA1 and BRCA2 associated risk have focused on a candidate gene approach, based on knowledge of gene functions, or the development of large genome wide association studies. In this study, researchers evaluated 24 single nucleotide polymorphisms tagged to 14 candidate genes derived through a novel approach that analysed gene expression differences to prioritize candidate modifier genes for association studies. [Breast Cancer Res] Mena Deficiency Delays Tumor Progression and Decreases Metastasis in Polyoma Middle-T Transgenic Mouse Mammary Tumors The actin binding protein Mammalian enabled (Mena), has been implicated in the metastatic progression of solid tumors in humans. Here, researchers report the effects of Mena deficiency on tumor progression, metastasis and on normal mammary gland development. [Breast Cancer Res] Epigenetic Control of the Basal-Like Gene Expression Profile via Interleukin-6 in Breast Cancer Cells Researchers conclude that IL-6, whose methylation-dependent autocrine loop is triggered by the inactivation of p53, induces an epigenetic reprogramming that drives breast carcinoma cells towards a basal-like/stem cell-like gene expression profile.
