    TOP STORY

Specific Interactions between Smad Proteins and AP-1 Components Determine TGFβ-Induced Breast Cancer Cell Invasion
Researchers investigated whether specific members of the AP-1 transcription factor family determine transforming growth factor β (TGFβ) signaling specificity in breast cancer cell invasion. [Oncogene] Abstract

    LABORATORY RESEARCH

ECM Microenvironment Regulates Collective Migration and Local Dissemination in Normal and Malignant Mammary Epithelium
To test the importance of the extracellular matrix (ECM) in tumor cell dissemination, researchers cultured epithelium from primary human breast carcinomas in different ECM gels. They used basement membrane gels to model the normal microenvironment and collagen I to model the stromal ECM. [Proc Natl Acad Sci USA] Abstract

Biomarkers of Response to Akt Inhibitor MK-2206 in Breast Cancer
Researchers determined whether allosteric Akt inhibitor MK-2206 inhibits tumor growth, and PTEN/PIK3CA mutations confer MK-2206 sensitivity. [Clin Cancer Res] Abstract

miR-100 Suppresses IGF2 and Inhibits Breast Tumorigenesis by Interfering with Proliferation and Survival Signaling
Investigators detected downregulation of microRNA-100 (miR-100) in breast cancer cells, leading to an upregulation of the proliferation- and survival-promoting oncogene insulin-like growth factor 2 (IGF2). Stable overexpression of miR-100 strongly reduced IGF2 expression and inhibited tumor growth. [Oncogene] Abstract

CCL2/CCR2 Chemokine Signaling Coordinates Survival and Motility of Breast Cancer Cells through Smad3 and p42/44MAP Kinase Dependent Mechanisms
Scientists characterized an important role for CCL2/CCR2 chemokine signaling in regulating the intrinsic relationships between breast cancer cell motility and survival, with implications on the metastatic process. [J Biol Chem] Abstract | Full Article

Expression of Aldehyde Dehydrogenase 1 as a Marker of Mammary Stem Cells in Benign and Malignant Breast Lesions of Ghanaian Women
Aldehyde dehydrogenase 1 (ALDH1) expression was higher in stromal components of benign compared with cancerous lesions. Of the estrogen receptor-, progesterone receptor-, and human epidermal growth factor receptor 2-defined subtypes of breast cancer, expression of ALDH1 was highest in triple-negative breast cancer. [Cancer] Abstract

A Switch Role of Src in the Biphasic EGF Signaling of ER-Negative Breast Cancer Cells
Scientists investigated the molecular mechanisms underlying the biphasic epidermal growth factor (EGF) signaling in ER-negative breast cancer MDA-MB-231 and MDA-MB-436 cells, both of which express endogenous EGF receptor. [PLoS One] Full Article

Luteolin Exerts Anti-Tumor Activity through the Suppression of Epidermal Growth Factor Receptor-Mediated Pathway in MDA-MB-231 ER-Negative Breast Cancer Cells
Researchers investigated the inhibitory effect of luteolin on MDA-MB-231 estrogen receptor (ER) negative breast tumor growth both in vitro and in vivo. [Food Chem Toxicol] Abstract

Human Wharton’s Jelly Stem Cell Conditioned Medium and Cell-Free Lysate Inhibit Human Osteosarcoma and Mammary Carcinoma Cell Growth In Vitro and in Xenograft Mice
Researchers examined the influence of human Wharton’s jelly stem cell conditioned medium and cell-free lysate on two osteosarcoma cell lines in vitro and on human mammary carcinomas in immunodeficient mice. [J Cell Biochem] Abstract

    CLINICAL RESEARCH

Five-Year Results of Whole Breast Intensity Modulated Radiation Therapy for the Treatment of Early Stage Breast Cancer: The Fox Chase Cancer Center Experience
Researchers report the five-year outcomes using whole-breast intensity modulated radiation therapy for the treatment of early stage breast cancer. [Int J Radiat Oncol Biol Phys] Abstract

High Incidence of Instability and Loss of Heterozygosity in Three Loci in Breast Cancer Patients Receiving Chemotherapy: A Prospective Study
Scientists evaluated potential chemotherapy-induced microsatellite instability, loss of heterozygosity, loss of expression in mismatch repair proteins and associations with clinical findings in breast cancer patients, especially resistance to chemotherapy and/or development of other tumors in the four years following chemotherapy treatment. Effect of Obesity on Toxicity in Women Treated with Adjuvant Chemotherapy for Early-Stage Breast Cancer: A Systematic Review
Authors provide evidence regarding the role of dose modification of chemotherapy in obese women with breast cancer by systematically reviewing current literature regarding chemotherapy-induced toxicity rates in obese and non-obese women with early-stage breast cancer. [Breast Cancer Res Treat] Abstract

    SCIENCE NEWS
Cup of Herbal Tea Could Help Fight Breast Cancer
Scientists at Aston University in Birmingham and Russells Hall Hospital have undertaken tests of an extract from a common plant in Pakistan and proved that it kills cancer cells without damage to normal breast cells in laboratory conditions. [Aston University] Press Release

New Data from Phase III EMILIA Study Showed Genentech’s Trastuzumab Emtansine (T-DM1) Significantly Improved Survival of People with HER2-Positive Metastatic Breast Cancer
Genentech announced updated results from the Phase III EMILIA study which showed that T-DM1 significantly extended the lives of people with HER2-positive metastatic breast cancer compared to the combination of lapatinib and Xeloda®. [Genentech, Inc.] Press Release
