Synthetic Lethality of PARP and Nicotinamide Phosphoribosyltransferase Inhibition in Triple-Negative Breast Cancer Cells
PARP inhibitors have been proposed as a potential targeted therapy for patients with triple-negative breast cancers. To better understand the mechanisms that determine the response to PARP inhibitors, researchers investigated whether enzymes involved in metabolism of the PARP substrate, β-NAD+, might alter the response to a clinical PARP inhibitor. LABORATORY RESEARCH

Major Histocompatibility Complex Class II+ Invariant Chain Negative Breast Cancer Cells Present Unique Peptides that Activate Tumor-Specific T Cells from Breast Cancer Patients
Researchers demonstrated that the major histocompatibility complex (MHC) class II-associated invariant chain, Ii, regulates the repertoire of tumor peptides presented by MHC class II+ breast cancer cells and identified novel immunogenic MHC II-restricted peptides that are potential therapeutic reagents for cancer patients. Reversal by RARα Agonist Am580 of c-Myc-Induced Imbalance in RARα/RARγ Expression during MMTV-Myc Tumorigenesis
The effects of c-Myc on retinoic acid receptor (RAR) isotype expression were evaluated in normal mouse mammary epithelium, mammary tumor cells obtained from the mouse mammary tumor virus (MMTV)-Myc transgenic mouse model as well human normal immortalized breast epithelial and breast cancer cell lines. The in vivo effect of the RARα-selective agonist Am580 was examined in the MMTV-Myc mouse model of mammary tumorigenesis. [Breast Cancer Res] Abstract | Full Article

Antagonists of Growth Hormone-Releasing Hormone Suppress In Vivo Tumor Growth and Gene Expression in Triple Negative Breast Cancers
Scientists evaluated the effects of a modern antagonistic analog of growth hormone-releasing hormone on tumor growth and on expression of inflammatory cytokine genes in two models of human triple negative breast cancers. [Oncotarget] Abstract

Ceramide Glycosylation by Glucosylceramide Synthase Selectively Maintains the Properties of Breast Cancer Stem Cells
Researchers report that ceramide glycosylation catalyzed by glucosylceramide synthase, that is enhanced in breast cancer stem cells (BCSCs) but not in normal mammary epithelial stem cells, maintains tumorous pluripotency of BCSCs. [J Biol Chem] Abstract | Full Article

Hypoxia Regulates CD44 and Its Variant Isoforms through HIF-1α in Triple Negative Breast Cancer
Researchers identified hypoxia inducible factor-1α (HIF-1α) as a regulator of CD44 that increased the number of CD44 molecules and the percentage of CD44 positive cells expressing variant exons v6 and v7/8 in breast cancer cells under hypoxic conditions. [PLoS One] Full Article

IPH-926 Lobular Breast Cancer Cells Harbor a p53 Mutant with Temperature-Sensitive Functional Activity and Allow for Profiling of p53-Responsive Genes
Profiling of p53-responsive genes has been carried out in different cellular models, most of which involved genetic modifications or cytotoxic stimulation. Scientists report on the utilization of IPH-926 human lobular breast cancer cells for the profiling of p53-responsive genes using a novel approach without such modifications. [Lab Invest] Abstract

The Novel Resveratrol Analogue HS-1793 Induces Apoptosis via the Mitochondrial Pathway in Murine Breast Cancer Cells
Researchers investigated the effect of HS-1793 as a new resveratrol (3,4′,5 tri-hydroxystilbene) analog on apoptosis via the mitochondrial pathway in murine breast cancer cells. [Int J Oncol] Abstract

Anti-Apoptotic Effect of Claudin-1 on TNF-α-Induced Apoptosis in Human Breast Cancer MCF-7 Cells
Researchers reported on the elevated expression of claudin-1 in human breast cancer MCF-7 cells under tumor necrosis factor-α (TNF-α) treatment. [Tumour Biol] Abstract

    CLINICAL RESEARCH

Clinicopathological Differences between Breast Cancer in Patients with Primary Metastatic Disease and Those Without: A Multicenter Study
Researchers suggested that receptor expression and histological type play a minor role in the risk for metastasis in patients with stage T3 and T4 disease when compared to patients with T1 primary metastatic breast cancer (pmBC) tumors. On initial diagnosis, lobular histology and luminal B positivity in T1 pmBC were determined to be risk factors for primary metastatic disease. [Eur J Cancer] Abstract

Long-Term Changes in Pulmonary Function after Incidental Lung Irradiation for Breast Cancer: A Prospective Study with Seven-Year Follow-Up
Investigators evaluated late pulmonary function changes after incidental pulmonary irradiation for breast cancer. [Int J Radiat Oncol Biol Phys] Abstract

The Interaction between ER and NFκB in Resistance to Endocrine Therapy
Authors provide an overview of the mechanisms of action of both transcription factors and focus on the current knowledge explaining how estrogen receptor (ER) and NFκB affect each other’s activity and how this cross-talk might contribute to the development of an endocrine resistance phenotype in breast cancer cells. [Breast Cancer Res] Abstract | Full Article

The Role of Cancer Stem Cells in Breast Cancer Initiation and Progression: Potential Cancer Stem Cell-Directed Therapies
This review summarizes the role of cancer stem cells (CSCs) in breast cancer recurrence and resistance and discusses current attempts of CSC targeting. [Oncologist] Abstract
