Hunk Negatively Regulates c-myc to Promote Akt-Mediated Cell Survival and Mammary Tumorigenesis Induced by Loss of Pten
Researchers report that Hormonally up-regulated neu-associated kinase (Hunk) is required for c-myc suppression and mammary tumorigenesis induced by phosphatase and tensin homolog (Pten) deletion in mice. Together, their findings establish a prosurvival function for Hunk in tumorigenesis, define an essential mechanism by which Akt suppresses c-myc-induced apoptosis, and identify Hunk as a previously unrecognized link between the Akt and c-myc oncogenic pathways. LABORATORY RESEARCH

Functional Cooperation of miR-125a, miR-125b, and miR-205 in Entinostat-Induced Downregulation of erbB2/erbB3 and Apoptosis in Breast Cancer Cells
Researchers demonstrated that entinostat specifically induces expression of microRNA (miR)-125a, miR-125b, and miR-205, which act in concert to downregulate erbB2/erbB3 in breast cancer cells. [Cell Death Dis] Full Article

Nox4-Dependent Reactive Oxygen Species Modulation by Amino Endoperoxides to Induce Apoptosis in Cancer Cells
The authors describe a one-pot strategy to synthesize novel amino endoperoxides and their derivatives with good yields and stabilities. In vitro cell-based assays revealed that four out of the 14 amino endoperoxides selectively induce metastatic breast carcinoma cells but not normal breast cells to undergo apoptosis, in a dose-dependent manner. EpCAM Expression Varies Significantly and Is Differentially Associated with Prognosis in the Luminal B HER2+, Basal-Like, and HER2 Intrinsic Subtypes of Breast Cancer
To investigate the association between epithelial cell adhesion molecule (EpCAM) expression and prognosis in the intrinsic subtypes of breast cancer, the authors performed immunohistochemical studies on a tissue microarray encompassing a total of 1365 breast cancers with detailed clinicopathological annotation and outcomes data. [Br J Cancer] Abstract

KISS1R Induces Invasiveness of Estrogen Receptor-Negative Human Mammary Epithelial and Breast Cancer Cells
Researchers report that either kisspeptin-10 treatment of estrogen receptor (ER)α-negative non-malignant mammary epithelial MCF10A cells or expression of KISS1R in MCF10A cells induced a mesenchymal phenotype and stimulated invasiveness. Similarly, exogenous expression of KISS1R in ERα-negative SKBR3 breast cancer cells was sufficient to trigger invasion and induced extravasation in vivo. [Endocrinology] Abstract

Syndecan Binding Protein (SDCBP) Is Overexpressed in Estrogen Receptor Negative Breast Cancers, and Is a Potential Promoter for Tumor Proliferation
SDCBP, an adapter protein containing PDZ domains, contributes to the tumorigenicity and metastasis of many malignant tumors, such as malignant melanoma. Scientists examined the expression profile of SDCBP in breast cancer (BCa) and its role in tumor cell proliferation, and then exploring its value in the targeted treatment of BCa. BRG1 Is a Prognostic Marker and Potential Therapeutic Target in Human Breast Cancer
Researchers explored the role of BRG1 in human breast cancer pathogenesis. Using tissue microarray and immunohistochemistry, they evaluated BRG1 staining in 437 breast cancer specimens and investigated its role in breast cancer cell proliferation, migration and invasion. Targeting Heat Shock Proteins by Phenethyl Isothiocyanate Results in Cell-Cycle Arrest and Apoptosis of Human Breast Cancer Cells
Researchers aimed to determine the effect of natural isothiocyanate (phenethyl isothiocyanate) on different heat shock proteins and heat shock factor 1 in two breast cancer cell lines, namely breast adenocarcinoma MCF-7 (with wild type p53) and highly metastatic breast cancer cell MDA-MB-231 (with mutated p53). [Nutr Cancer] Abstract

    CLINICAL RESEARCH

Large-Scale Genotyping Identifies 41 New Loci Associated with Breast Cancer Risk
The authors report a meta-analysis of nine genome-wide association studies, including 10,052 breast cancer cases and 12,575 controls of European ancestry, from which they selected 29,807 SNPs for further genotyping. They identified SNPs at 41 new breast cancer susceptibility loci at genome-wide significance. [Nat Genet] Abstract | Press Release

Enrichment of Regulatory T Cells in Invasive Breast Tumor Correlates with the Upregulation of IL-17A Expression and Invasiveness of the Tumor
Researchers found that regulatory T (Treg) and Th17 cells can affect disease progression by Treg-cell-mediated suppression of the effector T-cell response, as indicated by a decrease in the proliferation of T cells isolated from PBMCs of invasive ductal carcinoma of the breast patients and induction of angiogenic factors by IL-17-producing Th17. [Eur J Immunol] Abstract

Interferon-Induced Protein with Tetratricopeptide Repeats 1 (IFIT1) as a Prognostic Marker for Local Control in T1-2 N0 Breast Cancer Treated with Breast-Conserving Surgery and Radiation Therapy (BCS + RT)
Researchers evaluated the prognostic significance of IFIT1 protein expression in patients with breast cancer treated with BCS + RT. A tissue microarray was constructed with specimens from 282 women with node-negative, early-stage (I/II) breast cancer who were treated with BCS + RT. Immunohistochemistry was used to stain for the IFIT1 protein. [Breast J] Abstract

    REVIEWS

The Role of Intratumoral and Systemic IL-6 in Breast Cancer
The authors review the associations between IL-6 and breast cancer ranging from in vitro cell culture studies to clinical studies, covering the role of IL-6 in controlling breast cancer cell growth, regulation of cancer stem cell renewal, as well as breast cancer cell migration. [Breast Cancer Res Treat] Abstract

A Bi-Faceted Role of Estrogen Receptor Beta in Breast Cancer
In clinical studies and most often in cell culture studies, the pro-growth and pro-survival activity of estrogen receptor (ER) beta occurs in ER alpha negative breast cancer tissue and cells. This bi-faceted role of ER beta is reviewed. [Endocr Relat Cancer] Full Article
