    TOP STORY
First Large-Scale Proteogenomic Study of Breast Cancer Provides Insight into Potential Therapeutic Targets
Building on data from The Cancer Genome Atlas project, a multi-institutional team of scientists has completed the first large-scale “proteogenomic” study of breast cancer, linking DNA mutations to protein signaling and helping pinpoint the genes that drive cancer. [Press release from the Baylor College of Medicine discussing online prepublication in Nature] Press Release | Abstract

    LABORATORY RESEARCH

Dormant Breast Cancer Micrometastases Reside in Specific Bone Marrow Niches That Regulate Their Transit to and from Bone
Using real-time in vivo microscopy of bone marrow in a breast cancer xenograft model, researchers showed that dormant and proliferating breast cancer cells (BCCs) occupy distinct areas, with dormant BCCs predominantly found in E-selectin– and stromal cell–derived factor 1–rich perisinusoidal vascular regions. SnoN Antagonizes the Hippo Kinase Complex to Promote TAZ Signaling during Breast Carcinogenesis
Investigators observed previously that the intracellular localization and expression levels of SnoNare sensitive to cell density, suggesting that it may crosstalk with Hippo signaling. Here they report that SnoN interacts with multiple components of the Hippo pathway to inhibit the binding of Lats2 to TAZ and the subsequent phosphorylation of TAZ, leading to TAZ stabilization. [Dev Cell] Abstract

Functional Interaction of Histone Deacetylase 5 (HDAC5) and Lysine-Specific Demethylase 1 (LSD1) Promotes Breast Cancer Progression
The authors report that expression of HDAC5 and LSD1 proteins were positively correlated in human breast cancer cell lines and tissue specimens of primary breast tumors. [Oncogene] Abstract

Next-Generation Proteasome Inhibitor MLN9708 Sensitizes Breast Cancer Cells to Doxorubicin-Induced Apoptosis
Scientists used eight human breast cancer cell lines, which represent the major molecular subtypes of breast cancer, to validate the cytotoxic effects of MLN9708, alone and in combination with doxorubicin (Dox). They found that MLN9708 had cytotoxic effects, induced autophagy and MKP-1 expression, and enhanced Dox-induced apoptosis in these cell lines. [Sci Rep] Full Article

MUC1-C Stabilizes MCL-1 in the Oxidative Stress Response of Triple-Negative Breast Cancer Cells to BCL-2 Inhibitors
Researchers showed that targeting the oncogenic Mucin 1 (MUC1) C-terminal subunit (MUC1-C) in triple-negative breast cancer cells with silencing or pharmacologic inhibition with GO-203 is associated with downregulation of myeloid cell leukemia-1 (MCL-1) levels. [Sci Rep] Full Article

Long Intergenic Non-Coding RNA APOC1P1-3 Inhibits Apoptosis by Decreasing α-Tubulin Acetylation in Breast Cancer
Researchers found that 224 long non-coding RNAs (lncRNAs) were upregulated in breast cancer, whereas 324 were downregulated. The lincRNA-APOC1P1-3 was overexpressed in breast cancer, which was related to tumor size and hypomethylation in its promoter region. [Cell Death Dis] Full Article

    CLINICAL RESEARCH

Treatment Adherence and Its Impact on Disease-Free Survival in the Breast International Group 1-98 Trial of Tamoxifen and Letrozole, Alone and in Sequence
To investigate adherence to endocrine treatment and its relationship with disease-free survival in the Breast International Group 1-98 clinical trial. [J Clin Oncol] Abstract

Phase I Study of Pemetrexed with Sorafenib in Advanced Solid Tumors
To determine if combination treatment with pemetrexed and sorafenib is safe and tolerable in patients with advanced solid tumors [Oncotarget] Full Article

Bone Metastasis: The Importance of the Neighbourhood
The authors discuss the role of different bone cell types in supporting disseminated tumor cell dormancy and reactivation, and highlight the new opportunities this provides for targeting the bone microenvironment to control dormancy and bone metastasis. [Nat Rev Cancer] Abstract
