TOP STORYMetastatic Breast Cancer Cells Use Hedgehog to ‘Evilize’ Docile Neighbors Researchers showed that metastatic breast cancer cells signal neighboring cells in ways that allow otherwise anchored cells to metastasize. They pinpoint a promising link in the chain of signaling that, when broken, could reduce the metastatic potential of the disease. PUBLICATIONS(Ranked by impact factor of the journal)LABORATORY RESEARCHUsing a mouse model of spontaneous metastatic breast cancer, the authors investigated the molecular mediators of metastatic competence within a heterogeneous primary tumor and how these cells then manipulated their epithelial-mesenchymal plasticity during the metastatic process. [Sci Signal] Full Article To investigate phenotypic and genotypic alterations before and after bone metastasis, scientists conducted genome-wide mRNA profiling and DNA exon sequencing of two cell lines derived from a mouse xenograft model. [Sci Rep] Full Article TRPC5-Induced Autophagy Promotes Drug Resistance in Breast Carcinoma via CaMKKβ/AMPKα/mTOR Pathway Researchers demonstrated that adriamycin up-regulated both levels of TRPC5 and autophagy, and that the increase in autophagy was mediated by TRPC5 in breast cancer cells. Blockade of TRPC5 or autophagy increased the sensitivity to chemotherapy in vitro and in vivo. [Sci Rep] Full Article Molecular Characterization of Breast Cancer Cell Lines through Multiple Omic Approaches The authors characterized a large, commonly employed panel of breast cancer cell lines obtained from the American Type Culture Collection to enable researchers to make more informed decisions in selecting cell lines for specific studies. [Breast Cancer Res] Full Article Researchers report that in non-metastatic, estrogen receptor-positive breast cancer cells, induced mixed-lineage kinase 3 (MLK3) expression robustly upregulates the oncogenic transcription factor, FOS-related antigen-1 (FRA-1), which is accompanied by elevation of matrix metalloproteinases (MMPs), MMP-1 and MMP-9. [Oncogenesis] Full Article Proline-Rich Homeodomain Protein (PRH/HHEX) Is a Suppressor of Breast Tumor Growth To determine the consequences of PRH loss of function in breast cancer cells, investigators generated inducible PRH depletion in MCF-7 cells. They showed that PRH depletion resulted in increased MCF-7 cell proliferation in part at least due to increased vascular endothelial growth factor signaling. [Oncogenesis] Full Article | Press Release Scientists unveiled ten novel p53 target candidates that were up-regulated after the induction of p53 in wild-type cells. Their expressions were also high in breast invasive carcinoma tissues with wild-type p53. [Oncotarget] Full Article A Systematic Evaluation of Factors Affecting Extracellular Vesicle Uptake by Breast Cancer Cells By displaying various targeting peptides on the EV surface, investigators observed that although displaying GE11 on extracellular vesicles (EVs) modestly increased uptake by MCF-7 cells, NPY display had no effect on uptake by the same cells. In contrast, NTS and uPA display reduced EV uptake by MDA-MB-231 cells. [Tissue Eng Part A] Abstract Researchers compared estrogenic activity of bisphenol A to 6 bisphenol analogues in three human breast cancer cell lines. Estrogenicity was assessed by cell growth in an estrogen receptor-mediated cell proliferation assay, and by the induction of estrogen response element-mediated transcription in a luciferase assay. [Toxicol Sci] Abstract | Full Article The authors investigated the role of BDNF on the tumor cell microenvironment, namely, the cancer cell-endothelial cell interaction of TNBC cells. [PLoS One] Full Article CLINICAL RESEARCHIn this multicenter, Phase III, randomized, controlled trial, scientists enrolled patients aged 18 years or older from 129 UK centres who had histologically confirmed node-positive or high-risk node-negative operable breast cancer, had undergone complete excision, and were due to receive adjuvant chemotherapy. REVIEWS27-Hydroxycholesterol: A Novel Player in Molecular Carcinogenesis of Breast and Prostate Cancer The authors highlight recent findings pertaining to the role of 27 hydroxycholesterol in breast cancer and PCa and delineates the signaling mechanisms involved in the governing of different facets of tumor biology, that include tumor cell proliferation, epithelial-mesenchymal transition, as well as tumor cell invasion, migration, and metastasis. POLICY NEWSTexas Has Sanctioned Unapproved Stem Cell Therapies. Will It Change Anything? Texas Governor Greg Abbott signed a bill allowing clinics and companies in the state to offer people unproven stem cell interventions without the testing and approval required under federal law. Like the “right to try” laws that have sprung up in more than 30 states, the measure is meant to give desperately ill patients access to experimental treatments without oversight from the U.S. Food and Drug Administration. [ScienceInsider] Editorial Empty Rhetoric over Data Sharing Slows Science Government agencies lack the funds to build platforms for data sharing and resist taking responsibility for such infrastructure. They may hope that universities will host data, but the development of institutional repositories is patchy, and to rely on them is effectively to discourage common data standards and curation.
