TOP STORYResearchers define the lipogenic enzyme acetyl-CoA carboxylase (ACC) 1 as a key player in breast cancer metastasis. ACC1 phosphorylation was increased in invading cells both in murine and human breast cancer, serving as a point of convergence for leptin and transforming growth factor β signaling. PUBLICATIONS(Ranked by impact factor of the journal)Scientists showed that miR-31 is highly expressed in mammary stem cell (MaSC)-enriched mammary basal cell population and in mammary tumors, and is regulated by NF-κB signaling. They demonstrated that miR-31 promotes mammary epithelial proliferation and MaSC expansion at the expense of differentiation in vivo. [Nat Commun] Full Article Protein Acyltransferase DHHC3 Regulates Breast Tumor Growth, Oxidative Stress and Senescence Investigators showed that the protein palmitoyltransferase DHHC3 is upregulated in malignant and metastatic human breast cancer. Elevated expression of DHHC3 correlated with diminished patient survival in breast cancer and six other human cancer types. ZDHHC3 ablation in human MDA-MB-231 mammary tumor cell xenografts reduced the sizes of both the primary tumor and metastatic lung colonies. [Cancer Res] Abstract The normal adult human female mammary gland is a bilayered structure consisting of an outer basal layer and two readily distinguished subsets of cells within the inner luminal layer. The authors present a validated methodology for undertaking large-scale multi-parameter mass cytometric analyses of these cell types at single-cell resolution. [Cell Rep] Full Article | Graphical Abstract By utilizing cell proliferation assays, flow cytometry, immunocytochemistry, and cell migration/invasion assays, researchers examined the potentially tumorigenic activity of perfluorooctanesulfonic acid (PFOS) in MCF-10A breast cell line. [Arch Toxicol] Full Article To identify the role and to explore the mechanism of extracellular 5′-nucleotidase in human breast cancer growth, CD73 expression was measured firstly in breast cancer tissues and cell lines, and then interfered with or over-expressed by recombinant lentivirus in cell lines. [Int J Cancer] Abstract Scientists present the first demonstration that GBP2 inhibits mitochondrial fission and cell metastasis in breast cancer cells both in vitro and in vivo. They demonstrated that GBP2 binds directly to dynamin-related protein 1 (Drp1). Elimination of Drp1 by shRNA or Mdivi-1 (a Drp1-specific inhibitor) suppressed GBP2’s regulatory function. [Cell Death Dis] Full Article Scientists report the effects on apoptosis and autophagy in human triple-negative breast cancer cell lines after treatment with cantharidin and attempt to elucidate the underlying mechanisms. [Cell Physiol Biochem] Full Article Investigators showed that the Notch-4 intracellular domain (Int3) activates NF-κB in HC11 cells in absence of Rbpj through an association with the IKK signalosome. Int3 induced the canonical NF-κB activity and P50 phosphorylation in HC11 cells without altering the NF-κB2 pathway. [Sci Rep] Full Article The authors showed that Fasudil, a ROCK inhibitor, inhibited the migration of MDA-MB 231 and A549 cells, without altering the viability of these cells at the concentration of 10 μM, modified tumor cell morphology, with disorganization of stress fibers and promoted activation of the canonical-Wnt/beta-catenin pathway. [Sci Rep] Full Article Researchers investigated the mechanisms underlying the changes in gene expression patterns associated with the cross-talk between breast cancer cells and the stroma. Breast cancer-associated fibroblasts-secreted factors but also stromal content of the tumors remarkably activated specific genes characterized by a DNA methylation pattern: hypermethylation at transcription start site and shore regions. INDUSTRY NEWSResearcher Lands $450K Grant to Study Gut Bacteria’s Role in Breast Cancer University of Virginia researcher Melanie Rutkowski can envision a day when doctors prescribe a specific diet to prevent the spread of breast cancer. A day when doctors could identify women at high risk for breast tumors just by examining the bacteria in their guts. And now the Susan G. Komen organization has awarded her $450,000 to fund pioneering research that could make that happen. [Uiversity of Virginia] Press Release Tolero Pharmaceuticals, Inc. announced preclinical data supporting development of TP-1287 and TP-3654 in triple negative breast cancer. Key findings from cell culture models suggest that dual therapy with alvocidib and TP-3654 showed a 100% reduction in Myc protein expression. Combining TP-1287 and TP-3654 resulted in a 58.3% reduction of tumor growth in a xenograft model. [Tolero Pharmaceuticals, Inc. (PR Newswire Association LLC.)] Press Release | |

POLICY NEWSKey Legislator Tells Trump Officials to Back Off on Proposed Overhead Spending Cap for NIH An influential legislator wants President Donald Trump’s administration and fellow Republicans to drop the notion of capping overhead costs on grants funded by the National Institutes of Health (NIH). [ScienceInsider] Editorial Bitter CRISPR Patent War Intensifies The long-running battle over US patents for CRISPR–Cas9 gene editing continues. The Broad Institute of Cambridge, Massachusetts, filed a fresh set of arguments with the US government to defend a key patent. [Nature News] Editorial China Announces Plans to Fast-Track Drug Approval China is overhauling its drug-approval system to let companies bring their treatments to market quicker and more easily. The Communist Party of China and the State Council, two of the country’s most authoritative bodies, announced plans to reduce the backlog of medicines awaiting approval by the China Food and Drug Administration (CFDA). Policies will also be introduced to boost the productivity of Chinese drugmakers and spur innovation in health care. [Nature News] Editorial
