PUBLICATIONS(Ranked by impact factor of the journal)IN VITROA single-step method for extracellular matrix (ECM) functionalization on poly(lactic-co-glycolic acid) microspheres using a novel compound, dopamine-conjugated poly(ethylene-alt-maleic acid), as a stabilizer during the preparation of microspheres was reported. The dopamine molecules on the surface of microspheres provided active sites for the conjugation of ECM in an aqueous solution. [Small] Abstract Ankylosing spondylitis (AS)/β-TCP scaffolds were used as a three-dimensional biomimetic culture system to mimic the bone microenvironment in vivo to determine the abnormal osteogenic differentiation of AS-MSCs. Scientists demonstrated that when cultured in HA/β-TCP scaffolds, AS-MSCs had a stronger osteogenic differentiation capacity than that of MSCs from healthy donors in vitro and in vivo. [Cell Death Dis] Full Article Researchers showed that culture-adapted hepatic stellate cells (HeSCs) expressed substantially higher levels of hepatocyte growth factor, matrix metalloproteinase-1 and chemokine ligand 2 than bone marrow MSCs (BM-MSCs). Both human BM-MSCs and HeSCs inhibited T cell proliferation by a shared indoleamine, 2,3 dioxygenase-dependent mechanism. [Stem Cells] Abstract Investigators compared various preconditioning regimens for both the quantitative and qualitative production of mesenchymal stem cell-derived extracellular vesicles, and their therapeutic efficacy for proangiogenic activity in vitro and cutaneous wound healing in vivo. [J Clin Med] Full Article The authors described a method by which mammalian cells were added to silk solution before assembly, and thereby got uniformly integrated between the formed microfibers. In the resulting 3D scaffold, the cells were highly proliferative and spread out more efficiently than when encapsulated in a hydrogel. Elongated cells containing filamentous actin and defined focal adhesion points confirmed proper cell attachment to the FN-silk. [Sci Rep] Full Article Nestin+ cells were isolated using flow cytometry by gating for CD45−Ter119−CD31− cells from the compact bone of Nestin-GFP transgenic mice, expressing GFP driven by the Nestin promoter. Colony-forming and proliferative curve assays were conducted to determine the proliferative capacity of these cells, while qRT-PCR was used to analyze the mRNA levels of relative chemokines and growth factors. [Stem Cell Res Ther] Abstract IN VIVO4D Anisotropic Skeletal Muscle Tissue Constructs Fabricated by Staircase Effect Strategy The authors translated a staircase defect into an effective bioengineering strategy by integrating fused deposition modeling with surface coating technique to investigate the effect of topographical cues on regulating behaviors of human mesenchymal stem cells toward skeletal muscle tissues. This integrated approach served to fabricate shape-specific, multiple dimensional, anisotropic scaffolds using different biomaterials. [Biofabrication] Abstract Chitosan based hydrogels were developed to host mesenchymal stem cells, since their paracrine action can therapeutically enhance the spinal cord regeneration, limiting the formation of a glial scar and reducing cell death at the injured site. An injectable and highly permeable chitosan-based hydrogel was fabricated having a rapid gelation upon temperature increase from 0 to 37°C. [Sci Rep] Full Article The authors injected rat MSC intravitreally in adult Lister Hooded rats of either sex after optic nerve crush. Retinal ganglion cell survival, axonal regeneration, and reconnection were analyzed 60 and 240-days after crush by immunohistochemistry for Tuj1, anterograde labeling with cholera-toxin B and by immunohistochemistry for nerve growth factor-induced gene A in the superior colliculus after a cycle of light deprivation-stimulation. However, MSC transplantation in vivo must overcome the barriers including the low survival rate of transplanted stem cells, limited targeting capacity, and low grafting potency; therefore, much effort is needed to increase the survival and activity of MSCs in vivo.
