Hierarchically Assembled Nanofiber Scaffold Guides Long Bone Regeneration by Promoting Osteogenic/Chondrogenic Differentiation of Endogenous Mesenchymal Stem Cells

A 3D vertically aligned nanofiber scaffold (VAS) was developed with long channels and large pore size. The pore of VAS toward the bone marrow cavity after transplantation enabled the scaffolds to recruit bone marrow MSCs from the bone marrow cavity to the defect area.

Glucocorticoid Activates STAT3 and NF-κB Synergistically with Inflammatory Cytokines to Enhance the Anti-inflammatory Factor TSG6 Expression in Mesenchymal Stem/Stromal Cells

Investigators found that glucocorticoid could enhance the expression of TSG6, a vital tissue repair effector molecule, in IFNγ and TNFα treated human umbilical cord-MSCs.

Brg1/PRMT5 Nuclear Complex Epigenetically Regulates FOXO1 in IPF Mesenchymal Progenitor Cells

Scientists hypothesized that a Brg1/PRMT5 nuclear complex epigenetically regulates critical nodes in idiopathic pulmonary fibrosis (IPF) mesenchymal progenitor cell self-renewal signaling networks.

Solvent-Cast 3D Printing with Molecular Weight Polymer Blends to Decouple Effects of Scaffold Architecture and Mechanical Properties on Mesenchymal Stromal Cell Fate

Scaffold modulus did not affect human MSC differentiation under osteogenic conditions. However, human MSC response was significantly affected by scaffold stiffness in chondrogenic media.

Sequential Gene Expression Analysis of Myelodysplastic Syndrome Transformation Identifies HOXB3 and HOXB7 As the Novel Targets for Mesenchymal Cells in Disease

Researchers described sequential molecular changes and identified biomarkers in MSC of myelodysplastic syndrome (MDS) transformation. MDS-MSC was further isolated and cultured in vitro to determine the potential diagnostic and prognostic value of the identified biomarkers.

Placenta-Derived Mesenchymal Stem Cells Protect Against Diabetic Kidney Disease by Upregulating Autophagy-Mediated SIRT1/FOXO1 Pathway

Scientists explored the renoprotective effect of placenta-derived MSCs and the potential mechanism of SIRT1/FOXO1 pathway-mediated autophagy in diabetic kidney disease.

The Optimized Priming Effect of FGF-1 and FGF-2 Enhances Preadipocyte Lineage Commitment in Human Adipose-Derived Mesenchymal Stem Cells

To improve safety and graft vitality and to reduce unwanted lineage differentiation, this study refined the methodology by priming adipose-derived MSCs into preadipocytes—unipotent, self-renewing cells.

MSCs Mediate Long-Term Efficacy in a Crohn’s Disease Model by Sustained Anti-Inflammatory Macrophage Programming via Efferocytosis

Researchers used SAMP-1/YitFc, a chronic and spontaneous murine model of small intestinal inflammation, to study the therapeutic effects and mechanism of action of human bone marrow-derived MSCs.

Tumor-Derived Small Extracellular Vesicles Facilitate Omental Metastasis of Ovarian Cancer by Triggering Activation of Mesenchymal Stem Cells

The impact of extracellular vesicles and miR-320a-activated adipose-derived MSCs on tumor growth and metastasis was assessed in subcutaneous and orthotopic ovarian cancer xenograft mouse models.

EPA Scraps Plan to End Mammal Testing by 2035

The US Environmental Protection Agency (EPA) has abandoned a controversial plan to phase out all use of mammals to test the safety of chemicals by 2035. The hard deadline—imposed in 2019 to accelerate toward nonanimal models such as “organs on a chip”—made EPA unique among US federal agencies.
