Deubiquitinase MYSM1 Is Essential for Normal Bone Formation and Mesenchymal Stem Cell Differentiation
The authors described the function of MYSM1 on mesenchymal stem cell maintenance and differentiation. In this report, they found that Mysm1−/− mice had a lower bone mass both in long bone and calvaria compared with their control counterpart. [Sci Rep] Full Article

Extracellular Vesicles Derived from Osteogenically Induced Human Bone Marrow Mesenchymal Stem Cells Can Modulate Lineage Commitment
The osteoinductive potential of osteogenically induced human bone marrow mesenchymal stem cells-extracellular vesicles (hBMSC-EVs) was examined. hBMSCs secreted negatively charged nanosized vesicles with EV-related surface markers. The yield of EVs over seven days was dependent on an osteogenic stimulus. [Stem Cell Reports] Full Article | Graphical Abstract

Soft Elasticity-Associated Signaling and Bone Morphogenic Protein 2 Are Key Regulators of Mesenchymal Stem Cell Spheroidal Aggregates
As BMP2 up-regulation precedes that of interleukin-1 (IL1)B expression during spheroid formation, researchers hypothesized that BMP2 signaling triggers IL1 signaling in mesenchymal stem cell spheroids. Contrary to expectations, BMP2 signaling decreased expression of IL1B and downstream genes in a SMAD6-dependent manner. [Stem Cells Dev] Abstract

Differential Regulation of SOX9 Protein during Chondrogenesis of Induced Pluripotent Stem Cells versus Mesenchymal Stromal Cells: A Shortcoming for Cartilage Formation
In search for shortcomings of induced pluripotent stem cell (iPSC) chondrogenesis, scientists investigated whether SOX9 protein was adequately regulated during multi-phase chondrogenic differentiation of two human iPSC lines in a comparable manner like during mesenchymal stromal cell chondrogenesis. [Stem Cells Dev] Abstract

MiR-144-3p Regulates Osteogenic Differentiation and Proliferation of Murine Mesenchymal Stem Cells by Specifically Targeting Smad4
Researchers found that miR-144-3p is down-regulated during osteoblast differentiation of C3H10T1/2 cells. Overexpression of miR-144-3p inhibited osteogenic differentiation, whereas inhibition of miR-144-3p reversed this process. [FEBS Lett] Abstract

Sirolimus Induces Apoptosis and Reverses Multidrug Resistance in Human Osteosarcoma Cells In Vitro via Increasing MicroRNA-34b Expression
Given the up-regulation of mTOR pathway in many chemoresistant cancers, the authors examined whether sirolimus, a first generation mTOR inhibitor, might induce human osteosarcoma (OS) cell apoptosis and increase the sensitivity of OS cells to anticancer drugs in vitro. [Acta Pharmacol Sin] Full Article

Hypoxia Inhibits Mesenchymal Stem Cell Proliferation through HIF1α-Dependent Regulation of P27
Mesenchymal stem cells were isolated from adult male Fischer-344 rats bone marrow and kept under hypoxia. Cell survival and proliferation were studied using MTT and CFSE assay which showed reduced proliferation rate with an arrest in G0/G1 phase of cell cycle using flow cytometry. [Mol Cell Biochem] Abstract

Bone Marrow Stromal/Stem Cell-Derived Extracellular Vesicles Regulate Osteoblast Activity and Differentiation In Vitro and Promote Bone Regeneration In Vivo
Scientists explored the role of bone marrow stromal cell-derived extracellular vesicles (EVs) in the regulation of osteoblast activity and bone regeneration. They isolated bone marrow stromal/stem cell-derived EVs through gradient ultracentrifugation and ultrafiltration, and tested the influence of the EVs on osteogenesis both in vivo and in vitro. Human Amniotic Mesenchymal Stem Cells Alleviate Lung Injury Induced by Ischemia and Reperfusion after Cardiopulmonary Bypass in Dogs
Investigators assessed the capacity of human amniotic MSC to ameliorate ischemia/reperfusion injury in a dog model of cardiopulmonary bypass. [Lab Invest] Abstract

Allogeneic Mesenchymal Stem Cells in Combination with Hyaluronic Acid for the Treatment of Osteoarthritis in Rabbits
The authors investigated the effects of intraarticular injection of allogeneic mesenchymal stem cells in an in vivo anterior cruciate ligament transection model of osteoarthritis in rabbits. [PLoS One] Full Article

Articular Cartilage: From Formation to Tissue Engineering
The authors summarize the current state of cartilage tissue engineering in the context of relevant biological aspects, such as the formation and growth of hyaline cartilage, its composition, structure and biomechanical properties. Special attention is given to materials development, scaffold designs, fabrication methods, and template–cell interactions, which are of great importance to the structure and functionality of the engineered tissue.
