    TOP STORY

Adaptable Hemodynamic Endothelial Cells for Organogenesis and Tumorigenesis

Researchers demonstrated that ‘reset’ vascular endothelial cells established an adaptive vascular niche that differentially adjusted and conformed to organoids and tumoroids in a tissue-specific manner.

ILC1 Drive Intestinal Epithelial and Matrix Remodelling

Investigators developed gut organoid cocultures with type-1 innate lymphoid cells (ILC1) to dissect the impact of their accumulation in inflamed intestines.

Transplantable Jejunal Mucosal Grafts Using Patient-Derived Organoids from Children with Intestinal Failure

Researchers construct autologous jejunal mucosal grafts using biomaterials from pediatric patients and show that patient-derived organoids can be expanded efficiently in vitro.

Model Transcriptional Hallmarks of Oncogenic KRAS Activation in Lung Epithelial Progenitor Cells

Scientists developed organoid systems from primary mouse and human induced pluripotent stem cell-derived lung epithelial cells to model early-stage lung adenocarcinoma.

Generation of Functional Murine Heart Organoids via FGF4 and Extracellular Matrix

Investigators developed a method to generate heart organoids from mouse embryonic stem cell-derived embryoid bodies.

    Establishment
and Differentiation of Long-Term Trophoblast Organoid Cultures from the Human Placenta

Scientists describe a protocol to isolate trophoblast from first-trimester human placentas that can be grown long term in a three-dimensional organoid culture system.

    YAP
and AP-1 Cooperate to Initiate Pancreatic Cancer Development from Ductal Cells in Mice

The authors clarify the unique consequences of YAP/TAZ activation in the ductal cell population of the pancreas by generating mice with pancreatic duct cell-specific, inducible knockouts of Lats1 and Lats2, the main kinases upstream of YAP/TAZ.

    Modeling
Tumor Development and Metastasis Using Paired Organoids Derived from Patients with Colorectal Cancer Liver Metastases

Researchers generated paired organoids derived from primary tumors and matched liver metastases in the same colorectal cancer patients.

Gustatory Sensory G-Protein GNAT3 Suppresses Pancreatic Cancer Progression in Mice

Compromised gustatory sensing, achieved by Gnat3 ablation, enhanced the CXCL1/2 – CXCR2 axis to alter the myeloid-derived suppressor cell population and promoted the progression of metastatic pancreatic ductal adenocarcinoma.

Human Cerebral Organoids Reveal Early Spatiotemporal Dynamics and Pharmacological Responses of UBE3A

Human cerebral organoids revealed a change from weak to strong UBE3A in neuronal nuclei within three weeks of culture. Angelman syndrome human induced pluripotent stem cell-derived organoids also exhibited early silencing of paternal UBE3A, with topoisomerase inhibitors partially rescuing UBE3A levels and calcium transient phenotypes.

Cancer Cell-Originated Lactate Promotes the Self-Renewal of Cancer Stem Cells in Patient-Derived Colorectal Cancer Organoids

Organoid models were employed to show that cancer stem cells (CSCs) and non-CSCs in colorectal cancer were characterized by distinct metabolic phenotypes.
    [Cancer Letters]

    Abstract

3D Co-Culture of Cancer-Associated Fibroblast with Oral Cancer Organoids

The three-dimensional organoid model was employed to investigate whether co-culture with paralleled cancer-associated fibroblasts promoted stem-like properties of oral squamous cell carcinoma.
