Modeling Sporadic Alzheimer’s Disease in Human Brain Organoids under Serum Exposure

Because the blood–brain barrier (BBB) leakage is a well-known risk factor for Alzheimer’s disease (AD), brain organoids were exposed to human serum to mimic the serum exposure consequence of BBB breakdown in AD patient brains.

Application of Human Liver Organoids as a Patient-Derived Primary Model for HBV Infection and Related Hepatocellular Carcinoma

The authors described a primary ex vivo hepatitis B virus (HBV)-infection model derived from healthy donor liver organoids after challenge with recombinant virus or HBV-infected patient serum.

Klotho Inhibits Neuronal Senescence in Human Brain Organoids

Investigators showed that human cortical neurons, derived from human pluripotent stem cells in 2D cultures or in cortical organoids, developed the typical hallmarks of senescent cells when maintained in vitro for prolonged periods of time, and that moderated upregulation or repression of endogenous Klotho expression in cortical organoids inhibited and accelerated senescence, respectively.

O-GlcNAc Transferase Ogt Regulates Embryonic Neuronal Development through Modulating Wnt/β-Catenin Signaling

Scientists showed that Ogt was highly expressed in embryonic brain, and Ogt depletion reduced the proliferation of embryonic neural stem cells and migration of new born neurons.

Response to Elexacaftor/Tezacaftor/Ivacaftor in Intestinal Organoids Derived from People with Cystic Fibrosis

The authors utilized an intestinal organoid approach to compare in vitro responses to two CFTR modulator drug combinations and to check potential inter-individual variability in therapeutic response to the triple combination.

    Abstract

Effect of ABT-263 on Intestinal Fibrosis in Human Myofibroblasts, Human Intestinal Organoids, and the Mouse Salmonella typhimurium Model

Researchers evaluated the proapoptotic and antifibrotic efficacy of several classes of BH-3 mimetics in two in vitro fibrogenesis models. The candidate molecule, ABT-263, was advanced to a 3D human intestinal organoid model.

Lapatinib and Poziotinib Overcome ABCB1-Mediated Paclitaxel Resistance in Ovarian Cancer

Investigators established in vitro paclitaxel-resistant ovarian cancer cell line and organoid models. Gene expression differences in resistant and sensitive lines were analyzed by RNA sequencing.

Kidney Organoids Generated from Erythroid Progenitors Cells of Patients with Autosomal Dominant Polycystic Kidney Disease

Erythroid progenitors were reprogrammed by an episomal vector into iPSCs, which were differentiated into renal tubular organoids and then stimulated by forskolin to induce cysts formation.

Refining Epilepsy Treatment with Brain Organoids and AI

Monash epilepsy researchers are on an Medical Research Future Fund (MRFF) grant to personalise epilepsy anti-seizure treatment with brain organoids and artificial intelligence. Enabling innovative research that cuts across multiple scientific and clinical disciplines in order to address urgent unmet health care needs is what the MRFF scheme was designed to do, and is doing exactly that with its recently announced funding for a project to help patients with epilepsy.
