Splicing Factor SRSF1 Promotes Pancreatitis and KRASG12D-Mediated Pancreatic Cancer

Investigators reported that the splicing factor SRSF1 was highly expressed in pancreatitis, pancreatic ductal adenocarcinoma (PDAC) precursor lesions, and tumors. Increased SRSF1 was sufficient to induce pancreatitis and accelerate KRASG12D-mediated PDAC.

Targeting lncRNA DDIT4-AS1 Sensitizes Triple Negative Breast Cancer to Chemotherapy via Suppressing of Autophagy

Scientists found that the lncRNA, DNA damage inducible transcript 4 antisense RNA1 (DDIT4-AS1), was highly expressed in triple-negative breast cancer cell lines and tissues due to H3K27 acetylation in the promoter region.

RAD51 Foci as a Biomarker Predictive of Platinum Chemotherapy Response in Ovarian Cancer

RAD51 and γH2AX nuclear foci were evaluated by immunofluorescence in high-grade serous ovarian cancer patient-derived cell lines, organoids, and formalin-fixed, paraffin-embedded tumor samples.

    Abstract

An Automation Workflow for High-Throughput Manufacturing and Analysis of Scaffold-Supported 3D Tissue Arrays

Scientists optimized an automation approach to engineer tumor-mimetic 3D microtissues in Scaffold-supported Platform for Organoid-based Tissues using a liquid handler and showed comparable within- and between-sample variation to standard manual manufacturing.

    Abstract

Self-Organized Yolk Sac-Like Organoids Allow for Scalable Generation of Multipotent Hematopoietic Progenitor Cells from Induced Pluripotent Stem Cells

The authors found that hiPSCs cocultured with stromal cells as spheroids could grow in a stirred bioreactor and develop into yolk sac-like organoids without the addition of exogenous factors.

AAV-Mediated Gene Augmentation Therapy of CRB1 Patient-Derived Retinal Organoids Restores the Histological and Transcriptional Retinal Phenotype

Crumbs homolog 1 (CRB1) patient-derived PSC were differentiated into CRB1 retinal organoids that showed diminished expression of variant CRB1 protein observed by immunohistochemical analysis.

Organotypic Human Lung Bud Microarrays Identify BMP-Dependent SARS-CoV-2 Infection in Lung Cells

Investigators described a high-throughput platform to generate self-organizing and commensurate human lung buds derived from hESCs cultured on micropatterned substrates.

Novel Organoid Construction Strategy for Non-Involuting Congenital Hemangioma for Drug Validation

Researchers reported a novel non-involuting congenital hemangioma (NICH) organoid system construction and optimization process. Both HE and immunohistological staining exactly matched NICH tissue.
