Microglial Over-Pruning of Synapses during Development in Autism-Associated SCN2A-Deficient Mice and Human Cerebral Organoids

Scientists established a microglia-incorporated human cerebral organoid model carrying an SCN2A protein-truncating mutation identified in children with autism spectrum disorder. They found that human microglia display increased elimination of post-synapse in cerebral organoids carrying the SCN2A mutation.

    Abstract

Epidermal Growth Factor Receptor (EGFR) Is a Target of the Tumor-Suppressor E3 Ligase FBXW7

Investigators used CRISPR base editors to introduce different F-box and WD repeat domain-containing 7 (FBXW7) hotspot mutations in human colon organoids.

MSCs Alleviate LPS-Induced Acute Lung Injury by Inhibiting the Proinflammatory Function of Macrophages in Mouse Lung Organoid-Macrophage Model

Lung tissues were collected from mice for mouse lung organoid construction. Alveolar macrophages derived from bronchoalveolar lavage and interstitial macrophages (IMs) derived from lung tissue were co-cultured, with novel matrigel-spreading lung organoids to construct an in vitro model of lung organoids–immune cells.

Cholangiocyte Organoids to Study Drug-Induced Injury

Researchers investigated the mechanisms of chlorpromazine (CPZ)-induced bile duct injury using advanced in vitro cholangiocyte cultures. Intrahepatic cholangiocyte organoids were driven into mature cholangiocyte-like cells which were exposed to CPZ under cholestatic or non-cholestatic conditions.

cDC2 Plasticity and Acquisition of a DC3-Like Phenotype Mediated by IL-6 and PGE2 in a Patient-Derived Colorectal Cancer Organoids Model

Investigators used their recently developed 3D dendritic cell (DC)-tumor co-culture system to investigate how patient-derived primary and metastatic colorectal cancer organoids modulated conventional DC type 2 (cDC2) phenotype and function.

Human Cytomegalovirus (CMV) Dysregulates Neurodevelopmental Pathways in Cerebral Organoids

Scientists investigated potential CMV pathogenetic mechanisms of fetal neural malformation using in vitro human cerebral organoids. Cerebral organoids were permissive to CMV replication, and infection dysregulated cellular pluripotency and differentiation pathways.

OrgaSegment: Deep-Learning Based Organoid Segmentation to Quantify CFTR Dependent Fluid Secretion

Investigators presented OrgaSegment, a MASK-RCNN based deep-learning segmentation model allowing for the segmentation of individual intestinal patient-derived organoids structures from bright-field images.

Revolutionizing Pancreatic Islet Organoid Transplants: Improving Engraftment and Exploring Future Frontiers

Hypoxia-induced necrosis and delayed revascularization attenuate organoid viability and functional capacity, alleviated by the integration of diverse cell types e.g., human amniotic epithelial cells and human umbilical vein endothelial cells to boost vascularization.
