    TOP STORY

Sequential Progenitor States Mark the Generation of Pancreatic Endocrine Lineages in Mice and Humans

Researchers generated novel mouse lines and combined them with various genetic tools to enrich all types of hormone+ cells for well-based deep single-cell RNA sequencing (scRNA-seq), and gene coexpression networks were extracted from the generated data for the optimization of high-throughput droplet-based scRNA-seq analyses.

Neddylation Pathway Alleviates Chronic Pancreatitis by Reducing HIF1α-CCL5-Dependent Macrophage Infiltration

Scientists demonstrated that global inactivation of the neddylation pathway by MLN4924 significantly exacerbated chronic pancreatitis. The increased M2 macrophage infiltration, mediated by the upregulated chemokine ligand 5 (CCL5), was responsible for the enhanced pancreatitis-promoting activity of MLN4924.

Pre-Existing Beta Cells but Not Progenitors Contribute to New Beta Cells in the Adult Pancreas

The authors indicated that pancreatic endocrine and exocrine progenitor cells did not contribute to the new beta-cell formation in the adult mouse pancreas under physiological conditions.

Critical Parameters to Improve Pancreatic Cancer Treatment Using Magnetic Hyperthermia: Field Conditions, Immune Response, and Particle Biodistribution

Several alternating magnetic field conditions were evaluated using 3D cell culture models loaded with magnetic nanoparticles to determine which conditions were producing a strong effect on the cell viability.

pH-Sensitive Nanoparticles Composed Solely of Membrane-Disruptive Macromolecules for Treating Pancreatic Cancer

Investigators proposed long-circulating, pH-sensitive nanoparticles composed solely of cellular membrane-disruptive molecules as a new approach for treating pancreatic cancer.

    Abstract

Roundabout Homolog 1 Inhibits Proliferation via the YY1-ROBO1-CCNA2-CDK2 Axis in Human Pancreatic Cancer

Scientists explored whether ROBO1 had effects on the proliferation of pancreatic cancer and its specific mechanism.

    Abstract

Natural Product Trienomycin A Is a STAT3 Pathway Inhibitor That Exhibits Potent In Vitro and In Vivo Efficacy against Pancreatic Cancer

Researchers identified the natural product Trienomycin A, an ansamycin antibiotic, as a potential inhibitor of the Signal Transducer and Activator of Transcription-3 (STAT3) pathway with potent activity against pancreatic cancer.

    Abstract

EphA2 Super-Enhancer Promotes Tumor Progression by Recruiting FOSL2 and TCF7L2 to Activate the Target Gene EphA2

The authors explored the function and mechanism of a new super-enhancer in various tumor cells. A new super-enhancer that exists in a variety of tumors named EphA2-Super-enhancer was found using multiple databases and further identified.

Role of TGF-β in Pancreatic Ductal Adenocarcinoma Progression and PD-L1 Expression

Scientists investigated the expression of non-SMAD-transforming growth factor-beta (TGF-β) signaling proteins in patient-derived tissues, cell lines, and an immunocompetent mouse model.

    Abstract

SOX9 Modulates Cancer Biomarker and Cilia Genes in Pancreatic Cancer

To identify genes regulated by SOX9, researchers performed siRNA knockdown of SOX9 followed by RNA-seq in PANC-1 cells, a human PDAC cell line.

    Abstract

Elevated Levels of Mitochondrial CoQ10 Induce ROS-Mediated Apoptosis in Pancreatic Cancer

Investigators demonstrated that delivery of oxidized Coenzyme Q10 increased mitochondrial Q-pool and was associated with an increase in reactive oxygen species (ROS) generation, effectuating anti-cancer effects in a pancreatic cancer model.
