POLQ Inhibition Elicits an Immune Response in Homologous Recombination–Deficient Pancreatic Adenocarcinoma via cGAS/STING Signaling

Using human and murine homologous recombination (HR)-deficient PDAC models, scientists found that polymerase theta knockdown was synthetically lethal in combination with mutations in HR genes such as BRCA1 and BRCA2 and the DNA damage repair gene.

Lycopene Ameliorates Islet Function and Down-Regulates the TLR4/MyD88/NF-ÎşB Pathway in Diabetic Mice and Min6 Cells

By using anti-diabetic metformin as a positive control, investigators demonstrated that lycopene treatment suppressed islet inflammation and apoptosis in both high-fat diet /streptozotocin-induced diabetic mice and in Min6 cells exposed to high glucose/palmitic acid-RAW264.7 conditioned medium.

Membrane Protease Prostasin Promotes Insulin Secretion by Regulating the Epidermal Growth Factor Receptor Pathway

Researchers studied whether epidermal growth factor receptor (EGFR) was a substrate of prostasin (PRSS8) in pancreatic β-cells and that PRSS8 modulated insulin secretion via EGF–EGFR signaling.

Protocol Development to Further Differentiate and Transition Stem Cell-Derived Pancreatic Progenitors from a Monolayer Into Endocrine Cells in Suspension Culture

Scientists developed a protocol to harvest and aggregate human pluripotent stem cells-derived pancreatic progenitors generated using a commercially available kit into near uniform spheroids and to further differentiate the cells toward an endocrine cell fate in suspension culture.

Adipocytes Promote Pancreatic Cancer Migration and Invasion through Fatty Acid Metabolic Reprogramming

Using an in vitro coculture model, it was shown that adipocytes promoted tumor progression, and an intricate metabolic network between pancreatic cancer cells and adipocytes was identified and elucidated.

p53 Inhibitor iASPP Is an Unexpected Suppressor of KRAS and Inflammation-Driven Pancreatic Cancer

Researchers reported iASPP, an inhibitor of p53, as a paradoxical suppressor of inflammation and oncogenic KRASG12D-driven pancreatic cancer tumorigenesis.

Phosphoproteomics Guides Effective Low-Dose Drug Combinations Against Pancreatic Ductal Adenocarcinoma

Using two-step sequential phosphopeptide enrichment, scientists generated a comprehensive phosphoproteome and proteome of nine PDAC cell lines, encompassing more than 20,000 phosphosites on 5,763 phospho-proteins, including 316 protein kinases.

Targeting Pancreatic Cancer with Novel Plumbagin Derivatives: Design, Synthesis, Molecular Mechanism, In Vitro and In Vivo Evaluation

Investigators synthesized a series of plumbagin derivatives and found that 2-(cyclohexylmethyl)-plumbagin inhibited the PI3K/Akt/mTOR signaling pathways, leading to cancer cell death under nutrient-deprived conditions.

An Increased Demand of FAD Synthesis in Differentiated and Stem Pancreatic Cancer Cells Is Accomplished by Modulating FLAD1 Gene Expression

The FAD synthesis process was evaluated in two PDAC cell lines: PANC-1 and PANC-1 derived cancer stem cells (CSCs), presenting the R273H mutation in the oncosuppressor p53, as well as MiaPaca2 and MiaPaca2 derived CSCs, presenting the R248W mutation in p53.

Pancreatic β-Cell Heterogeneity in Adult Human Islets and Stem Cell-Derived Islets

The authors summarize the heterogeneity in islet cells of the adult pancreas and those generated from stem cells. They also highlight the significance of this heterogeneity in health and disease conditions and how this can be used to design a stem cell-derived product for diabetes cell therapy.
