Excess Pancreatic Elastase Alters Acinar-β Cell Communication by Impairing the Mechano-Signaling and the PAR2 Pathways

Using high-throughput screening assays, the authors identified telaprevir as a potent pancreatic elastase inhibitor that could increase human and rodent β cell viability in vitro and in vivo, and improve glucose tolerance in insulin-resistant mice.

MSCs Deliver Hypoxia-Treated Mitochondria Reprogramming Acinar Metabolism to Alleviate Severe Acute Pancreatitis Injury

It was shown that mesenchymal stem cells (MSCs) delivered hypoxia-treated functional mitochondria to damaged pancreatic acinar cells (PACs) via extracellular vesicles, which reversed the metabolic function of PACs, maintained ATP supply, and exhibited an excellent injury-inhibiting effect.

Beta Cell Dysfunction Induced by Bone Morphogenetic Protein (BMP)-2 Is Associated with Histone Modifications and Decreased NeuroD1 Chromatin Binding

Mouse islets exposed to BMP-2 for 10 days showed impaired glucose-stimulated insulin secretion and beta cell proliferation, and BMP-2-induced beta cell dysfunction was associated with decreased expression of cell maturity and proliferation markers specific to the beta cell.

Raptor Levels Are Critical for β-Cell Adaptation to a High-Fat Diet in Male Mice

Researchers used mice with heterozygous deletion of raptor in β-cells to assess whether reduced mTORC1 function was critical for β-cell function in normal conditions, or during β-cell adaptation to high-fat diet.

    Abstract

SOCS3 Inhibits the Mesenchymal Stromal Cell Secretory Factor SDF-1-Mediated Improvement of Islet Function in Non-Obese Diabetic Mice

Stromal cell-derived factor 1 (SDF-1) protected islet cells from cytokine-induced apoptosis in vitro. Suppressor of cytokine signaling 3 (SOCS3)-knockout islets pretreated with SDF-1 were effective in reducing blood glucose in non-obese diabetic mice in vivo.

Synthetically Lethal Biomimetic Nutri-Hijacker Hitchhikes and Reprograms KRAS Mutation-Driven Metabolic Addictions for Pancreatic Ductal Adenocarcinoma Treatment

The authors proposed a biomimetic “Nutri-hijacker” with “Trojan horse” design to induce synthetic lethality in KRAS-mutated malignant cells by hitchhiking and reprogramming the metabolic addictions.

Exploring Switch II Pocket Conformation of KRAS(G12D) with Mutant-Selective Monobody Inhibitors

When expressed in cells as genetically encoded reagents, these monobodies engaged selectively with KRAS(G12D) and inhibited KRAS(G12D)-mediated signaling and tumorigenesis.

Inhibition of Phosphodiesterase 4D Suppresses mTORC1 Signaling and Pancreatic Cancer Growth

Inhibition of phosphodiesterase 4D and the elevation of cAMP levels blocked mTORC1 signaling via Raptor phosphorylation. Pancreatic cancer exhibited an upregulation of phosphodiesterase 4D expression, and high levels predicted the poor overall survival of patients with pancreatic cancer.

Features of Metabolism Associated Molecular Patterns in Pancreatic Ductal Adenocarcinoma

Using random forest classifier, probabilities of three patterns could be predicted on pancreatic cancer cell lines, which could be used to find vulnerable targets in response to both genetic and drug perturbation.
