    TOP STORY
Modified Vitamin D Shows Promise as Treatment for Pancreatic Cancer
A synthetic derivative of vitamin D was found by researchers to collapse the barrier of cells shielding pancreatic tumors, making this seemingly impenetrable cancer much more susceptible to therapeutic drugs. DIABETES & PANCREATITIS

Metabolic Inflexibility Impairs Insulin Secretion and Results In MODY-Like Diabetes in Triple FoxO-Deficient Mice
Researchers show that ablation of the three FoxO genes in mature β cells results in early-onset, maturity-onset diabetes of the young (MODY)-like diabetes, with abnormalities of the MODY networks Hnf4α, Hnf1α, and Pdx1. [Cell Metab] Abstract | Graphical Abstract

CUX1 – A Modulator of Tumor Aggressiveness in Pancreatic Neuroendocrine Neoplasms
Researchers aimed to characterize the impact of the oncogenic transcription factor CUX1 on proliferation, resistance to apoptosis and angiogenesis in murine and human pancreatic neuroendocrine neoplasms. Expression and function of CUX1 was analyzed using knockdown and overexpression strategies in Ins-1 and Bon-1 cells, xenograft models and a genetically engineered mouse model of insulinoma. [Endocr Relat Cancer] Abstract

SMAD4 Regulates Cell Motility through Transcription of N-Cadherin in Human Pancreatic Ductal Epithelium
Expression of N-cadherin on the surface of epithelial cells facilitates motility and invasion, and scientists demonstrate that knockdown of SMAD4 causes decreased N-cadherin expression, which results in diminished migration and invasion of human pancreatic ductal epithelial cells. [PLoS One] Full Article

Glucagon-Like Peptide-1 Stimulates Type 3 Iodothyronine Deiodinase Expression in a Mouse Insulinoma Cell Line
Researchers identified deiodinase (D3) expression in MIN6 cells derived from a mouse insulinoma cell line and examined the mechanisms regulating D3 expression in these cells. [Life Sci] Full Article

Increased Expression of ERp57/GRP58 Is Protective against Pancreatic Beta Cell Death Caused by Autophagic Failure
The mechanisms involved in the deterioration of beta cell function caused by autophagic failure have not yet been fully elucidated. To gain insight into its mechanisms, scientists compared the protein expression of islets from beta cell-specific Atg7-deficient mice and their controls. [Biochem Biophys Res Commun] Abstract

    PANCREATIC CANCER

A Hypusine-eIF5A-PEAK1 Switch Regulates the Pathogenesis of Pancreatic Cancer
Knockdown of eIF5A proteins in pancreatic ductal adenocarcinoma (PDAC) cells inhibited their growth in vitro and orthotopic tumor growth in vivo, whereas amplification of eIF5A proteins increased PDAC cell growth and tumor formation in mice. [Cancer Res] Abstract

A TrxR Inhibiting Gold(I) NHC Complex Induces Apoptosis through ASK1-p38-MAPK Signaling in Pancreatic Cancer Cells
To understand the biological function of gold(I) N-heterocyclic carbene (NHC) complexes in pancreatic ductal adenocarcinoma (PDAC), scientists used a recently published gold(I) NHC complex, MC3, and evaluated its anti-proliferative effect in four PDAC cell lines, determined by MTT and SRB assays. [Mol Cancer]

Curcumin Inhibits Cell Growth and Invasion through Up-Regulation of miR-7 in Pancreatic Cancer Cells
Scientists explored whether curcumin regulates miR-7, leading to the inhibition of cell growth, migration and invasion in pancreatic cancer cells. [Toxicol Lett] Abstract

Differential Modulation of Nicotine-Induced Gemcitabine Resistance by GABA Receptor Agonists in Pancreatic Cancer Cell Xenografts and In Vitro
In vitro exposure of cells to single doses or seven days of nicotine induced the protein expression of MMP-2, MMP-9 and EGR-1 and these responses were blocked by GABA. Baclofen downregulated the protein expression of GABA-B-receptors in xenograft tissues and in cells exposed to baclofen for seven days in vitro. [BMC Cancer] Abstract | Full Article

Thiazolidinediones and the Promise of Insulin Sensitization in Type 2 Diabetes
Based on critical appraisal of both preclinical and clinical studies, the authors discuss the prospect of harnessing the insulin sensitizing effects of PPARγ for more effective, safe, and potentially personalized treatments of type 2 diabetes. [Cell Metab] Abstract
