TOP STORYEGFR Signaling Controls Cellular Fate and Pancreatic Organogenesis by Regulating Apicobasal Polarity Scientists found that ligand-specific EGFR signaling via PI(3)K and Rac1 autonomously modulates apicobasal polarity to enforce the sequential control of morphogenesis and cell differentiation. PUBLICATIONS(Ranked by impact factor of the journal)DIABETES & PANCREATITISResearchers investigated lysine acetylation in mitochondrial proteins from INS-1E β cells and in proteins from human pancreatic islets after 24 hour palmitate exposure. [Sci Rep] Full Article The Serum Protein Renalase Reduces Injury in Experimental Pancreatitis The authors found that calcium ATPase 4b (PMCA4b) is expressed in both murine and human acinar cells and that a PMCA4b selective inhibitor worsens pancreatitis-induced injury and blocked the protective effects of renalase. [J Biol Chem] Abstract | Full Article Scientists investigated the generation mechanism of new beta cells by considering the compensation phase of type 2 diabetes mellitus. Pancreatic islet derived mesenchymal stem cells were isolated from adult rats and characterized. [J Cell Physiol] Abstract Investigators evaluated the use of miR-375 biomarker to assess the beta cytoprotective effect of phenylpropenoic acid glucoside, which was previously demonstrated to protect beta cells against various types of injury, and of exendin-4, which is an established antidiabetic drug. [PLoS One] Full Article PANCREATIC CANCERSurvival of Pancreatic Cancer Cells Lacking KRAS Function The authors modeled complete KRAS inhibition using CRISPR/Cas-mediated genome editing and demonstrated that KRAS is dispensable in a subset of human and mouse pancreatic ductal adenocarcinoma cells. [Nat Commun] Full Article Investigators showed that identical oncogenic drivers trigger pancreatic ductal adenocarcinoma originating from both ductal and acinar cells with similar histology, but with distinct pathophysiology and marker expression dependent on cell of origin. [Cell Rep] Full Article | Graphical Abstract FAM49B, a Novel Regulator of Mitochondrial Function and Integrity That Suppresses Tumor Metastasis Scientists investigated the pro-tumoral roles of mitochondrial dynamics and altered intracellular reactive oxygen species levels in pancreatic ductal adenocarcinoma. They identified family with sequence similarity 49 member B (FAM49B) as a mitochondria-localized protein that regulates mitochondrial fission and cancer progression. [Oncogene] Full Article SOX9 Activity Is Induced by Oncogenic Kras to Affect MDC1 and MCMs Expression in Pancreatic Cancer Researchers investigated the mechanistic link between sex determining region Y-box 9 (SOX9) and oncogenic Kras, studied biological function of SOX9, and identified SOX9-related genes and their clinical significance in patients with pancreatic ductal adenocarcinoma. [Oncogene] Abstract Through miRNA profiling of gemcitabine-resistant (GR) and parental PANC-1 cell lines, scientists found a consistent reduction of miR-3656 in GR PANC-1 cells. miR-3656 overexpression enhanced the antitumor effect of gemcitabine, whereas silencing of miR-3656 resulted in the opposite effect. POLICY NEWSIranian Scholar Sentenced to Death A judge in Tehran has ordered the death penalty for Iranian researcher Ahmadreza Djalali, according to his wife and diplomatic sources in Italy. Djalali is affiliated with the Karolinska Institute in Stockholm, Sweden, and the University of Eastern Piedmont in Novara, Italy. A resident of Sweden with his family, Djalali was arrested in April 2016 on an academic visit to Tehran and accused of “collaboration with a hostile government”. [Nature News] Editorial In Brazil, Researchers Struggle to Fend Off Deepening Budget Cuts With time and money running out, Brazilian scientists are turning up the pressure on the federal government to avoid a total collapse of the national science and technology funding system before the end of the year. Researchers delivered a petition with more than 82,000 signatures to congressional leaders in Brasília, demanding the reversal of deep budget cuts that have left research institutions struggling to pay even basic water and electricity bills.
