    SCIENCE NEWS

Supercomputing Research Opens Doors for Drug Discovery
Researchers adapted a widely used existing software to allow supercomputers such as Oak Ridge National Laboratory’s (ORNL) Jaguar to sift through immense molecular databases and pinpoint chemical compounds as potential drug candidates. The project team plans to put the computational development to work on ORNL supercomputers to look for chemicals that could treat prostate cancer. [Oak Ridge National Laboratory Press Release]

Could Alpha-2 Integrin be a Protein Predictor of Tumor Spread?
Researchers have generated data that lead them to suggest that decreased expression of the protein alpha-2 integrin is predictive of tumor dissemination to distant sites and decreased survival in individuals with either breast or prostate cancer. [Press release from Medical News Today discussing online prepublication in the Journal of Clinical Investigation]

    LABORATORY RESEARCH

Somatic Mutations throughout the Entire Mitochondrial Genome Are Associated with Elevated PSA Levels in Prostate Cancer Patients
Researchers sequenced the entire mitochondrial genome (16,569 bp) from 30 prospectively collected pairs of macrodissected cancerous and benign cells from prostate cancer patients and compared their genetic variability. [Am J Hum Genet]

Invasive Prostate Carcinoma Driven by c-Src and Androgen Receptor Synergy
Researchers analyzed over-expression of cellular Src (c-Src) in naïve mouse prostate epithelium and observed no change in tubule formation frequency or histological structure. [Cancer Res]

Tyramide Signal Amplification for Antibody-Overlay Lectin Microarray: A Strategy to Improve the Sensitivity of Targeted Glycan Profiling
Here, researchers describe a new Tyramide Signal Amplification (TSA) for Antibody-overlay Lectin Microarray procedure for sensitive profiling of glycosylation patterns. Researchers demonstrated that TSA increased the sensitivity of the microarray over 100 times for glycan profiling using the model protein prostate specific antigen. [J Proteome Res]

Toward an Automatic Method for Extracting Cancer- and Other Disease-Related Point Mutations from the Biomedical Literature
Researchers introduce a high-throughput computational method for the identification of relevant disease mutations in Pub-Med abstracts applied to prostate and breast cancer mutations. [Bioinformatics]

Leptin Signaling and Apoptotic Effects in Human Prostate Cancer Cell Lines
This research studied leptin-induced signaling and apoptosis in androgen sensitive (LNCaP, PC3/AR) and insensitive (PC3, DU145) prostate cancer cell lines. [Prostate]

Anti-Angiogenic Effects and Mechanism of Prazosin
Alpha1-adrenoceptors antagonists (doxazosin, terazosin, prazosin) are commonly prescribed for benign prostate hyperplasia and hypertension. Doxazosin and terazosin exhibit anti-angiogenic effects and apoptotic activities against multiple cell types and are potential preventive agents for prostate cancer. Prazosin induces apoptosis in three prostate cancer cell lines. Researchers hypothesized that prazosin, a more potent alpha1-adrenoceptor antagonist with a distinct mechanism, exhibits anti-angiogenic activity. [Prostate]

Elevated GnRH Receptor Expression Plus GnRH Agonist Treatment Inhibits the Growth of a Subset of Papillomavirus 18-Immortalized Human Prostate Cells
Engineered high-level GnRH receptor activation inhibits growth of a subset of papillomavirus-immortalized prostate cells. Elucidating mechanisms leading to clone-specific differences in cell surface GnRH receptor levels is a valuable next step in developing strategies to exploit prostate cell anti-proliferation using GnRH agonists. [Prostate]

    CLINICAL RESEARCH

Impact of Age at Diagnosis on Prostate Cancer Treatment and Survival
Older patients are more likely to have high-risk prostate cancer at diagnosis and less likely to receive local therapy. Indeed, underuse of potentially curative local therapy among older men with high-risk disease may in part explain observed differences in cancer-specific survival across age strata. [J Clin Oncol]

Regulation of HMGA1 Expression by microRNA296 Affects Prostate Cancer Growth and Invasion
In this study, researchers examined the functional relation between miR-296 and high mobility group AT-hook 1 (HMGA1) expression in several prostate cancer cell lines and a large prostate cancer cohort. [Clin Cancer Res]
