    TOP STORY
Definition of Molecular Determinants of Prostate Cancer Cell Bone Extravasation
Scientists described how metastatic advanced prostate cancer (PCa) cells breach bone marrow endothelium (BMEC) monolayers in a step-wise fashion under physiologic hemodynamic flow. PCa cells tethered and rolled on BMEC and then firmly adhered to and traversed BMEC via sequential dependence on E-selectin ligands and β1 and αVβ3 integrins. [Cancer Res] Abstract

    LABORATORY RESEARCH

Cell-Based Selection Provides Novel Molecular Probes for Cancer Stem Cells
Because few markers exist to identify and isolate pure cancer stem cells (CSC), researchers used cell-based systematic evolution of ligands by exponential enrichment to create DNA aptamers that can identify novel molecular targets on the surfaces of live CSC. Out of 22 putative DNA sequences, three bound to >90% and five bound to >15% of DU145 prostate cancer cells. [Int J Cancer] Abstract

Reduction of Pro-Tumorigenic Activity of Human Prostate Cancer-Associated Fibroblasts Using Dlk1 or SCUBE1
Researchers set out to reduce the pro-tumorigenic activity of patient cancer-associated fibroblasts (CAFs) using the Dlk1 and SCUBE1 molecules that they had previously identified in prostate development. Their hypothesis was that mesenchymally expressed molecules might reduce CAF pro-tumorigenic activity, either directly or indirectly. [Dis Model Mech] Abstract | Press Release

Anti-Proliferative Effects of Physiological Concentrations of Enterolactone in Models of Prostate Tumorigenesis
Scientists investigated the anti-proliferative effects of enterolactone in an in vitro model of prostate tumorigenesis at concentrations reported to occur in a range of male populations. [Mol Nutr Food Res] Abstract

Subcellular Localization of p44/WDR77 Determines Proliferation and Differentiation of Prostate Epithelial Cells
Investigators found that p44 localizes in the cytoplasm of prostate epithelial cells at the early stage of prostate development when cells are proliferating, and its nuclear translocation is associated with cellular and functional differentiation in adult prostate tissue. [PLoS One] Full Article

Human α2β1HI CD133+VE Epithelial Prostate Stem Cells Express Low Levels of Active Androgen Receptor
Researchers re-examined the expression patterns of androgen receptor (AR) within adult prostate epithelial differentiation using an optimized sensitive and specific approach examining transcript, protein and AR regulated gene expression. [PLoS One] Full Article

The Chicken Ovalbumin Upstream Promoter-Transcription Factor II Negatively Regulates the Transactivation of Androgen Receptor in Prostate Cancer Cells
Scientists explored a putative role of chicken ovalbumin upstream promoter-transcription factor II (COUP-TF II) in prostate cancers by investigating its effect on cell proliferation and a cross-talk between COUP-TF II and androgen receptor. [PLoS One] Full Article

Tumor-Associated Stromal Cells Expressing E-Prostanoid 2 or 3 Receptors in Prostate Cancer: Correlation with Tumor Aggressiveness and Outcome by Angiogenesis and Lymphangiogenesis
To clarify the detailed pathologic roles of prostaglandin E2 in prostate cancer tissues, the authors investigated the clinical significance and prognostic roles of the density of tumor-associated stromal cells expressing specific receptors for prostaglandin E2, termed “E-prostanoid 1-4 receptors.” [Urology] Abstract

    CLINICAL RESEARCH

Impact of the Introduction of a Robotic Training Programme on Prostate Cancer Stage Migration at a Single Tertiary Referral Centre
2511 consecutive patients treated with radical prostatectomy, with or without pelvic lymph node dissection, for prostate cancer (PCa) at a single tertiary care center were analyzed. Baseline patient characteristics and PCa risk distribution were compared according to treatment type (retropubic radical prostatectomy [RRP] vs robot-assisted radical prostatectomy [RARP]) in the overall population, as well as in three surgeons’ initial 50 RARP and three surgeons’ initial 50 RRP cases. [BJU Int] Abstract

Second to Fourth Digit Ratio and Prostate Cancer Severity
The authors investigated the ratio of the second to the fourth digit as a potential marker for prostate cancer severity. [Prostate Cancer Prostatic Dis] Abstract

Detect 7 times more prostate epithelial progenitors with ProstaCult

    REVIEWS
The Complexity of Prostate Cancer: Genomic Alterations and Heterogeneity
The clinical heterogeneity of prostate cancer can be partly explained by this underlying genetic heterogeneity, which has been observed between patients from different geographical and ethnic populations, different individuals within these populations, different tumor foci within the same patient, and different cells within the same tumor focus. [Nat Rev Urol] Abstract

    INDUSTRY NEWS

TEGH Supporter and Celebrated Entrepreneur W. Brett Wilson Buoys Robotic Prostate Cancer Program with a Healthy Million-Dollar Challenge during “Movember”
Toronto East General Hospital (TEGH) supporter, iconic Canadian investor, grateful former TEGH patient and prostate-cancer “graduate”, W. Brett Wilson, has donated $100,000 to TEGH, launching the TEGH W. Brett Wilson Gift Challenge. Wilson is challenging Canadians to match his commitment, to hit a target of one million, in support of the hospital’s revolutionary robotic prostate cancer program.
