    TOP STORY
Prostate Development Discovery Could Lead to New Treatments
Scientists discovered how the prostate gland develops for the first time, according to research. The team behind the investigation says the findings could open the door to the development of new therapies for the treatment of prostate cancer. [Press release from the University of York discussing online prepublication in Stem Cell Reports] Press Release | Full Article | Graphical Abstract

Cofilin Drives Cell Invasive and Metastatic Responses to TGF-ß in Prostate Cancer
Cofilin binding and severing of F-actin is controlled by Ser3 phosphorylation, but the contributions of this step to cell migration during invasion and metastasis of cancer cells are unclear. Researchers addressed the question in prostate cancer cells, including the response to TGF-ß, a critical regulator of migration. [Cancer Res] Abstract

Targeting Src-Mediated Tyr216 Phosphorylation and Activation of GSK-3 in Prostate Cancer Cells Inhibit Prostate Cancer Progression In Vitro and In Vivo
Scientists investigated the role of glycogen synthase kinase-3 (GSK-3) and the existence of an Akt-resistant pathway for GSK-3 activation in prostate cancer cells. [Oncotarget] Full Article

PSMA-Targeted Stably Linked “Dendrimer-Glutamate Urea-Methotrexate” as a Prostate Cancer Therapeutic
Researchers report the preclinical evaluation of a serum-stable dendrimer-based drug conjugate capable of actively targeting into prostate cancer cells, delivered through the prostate-specific membrane antigen (PSMA). [Biomacromolecules] Abstract

Prostate Cancer Bone Metastases Acquire Resistance to Androgen Deprivation via WNT5A-Mediated BMP-6 Induction
Researchers investigated the role of bone morphogenetic protein-6 (BMP-6) in the development of castration-resistant prostate cancer in the context of bone metastases. [Br J Cancer] Abstract

The cAMP Phosphodiesterase-4D7 (PDE4D7) Is Downregulated in Androgen-Independent Prostate Cancer Cells and Mediates Proliferation by Compartmentalizing cAMP at the Plasma Membrane of VCaP Prostate Cancer Cells
Scientists identified the differential regulation of the PDE4D7 isoform during prostate cancer progression and uncover a role in controlling prostate cancer cell proliferation. [Br J Cancer] Abstract

A Humanized Tissue-Engineered In Vivo Model to Dissect Interactions between Human Prostate Cancer Cells and Human Bone
Investigators introduce a tissue engineering approach to explore the interactions between human tumor cells and a humanized bone microenvironment. Scaffolds, seeded with human primary osteoblasts in conjunction with BMP7, were implanted into immunodeficient mice to form humanized tissue engineered bone constructs (hTEBCs) which consequently resulted in the generation of highly vascularized and viable humanized bone. At 12 weeks, PC3 and LNCaP cells were injected into the hTEBCs. [Clin Exp Metastasis] Abstract

Antiproliferative Activity and Induction of Apoptosis in PC-3 Cells by the Chalcone Cardamonin from Campomanesia adamantium (Myrtaceae) in a Bioactivity-Guided Study
Ethanol extracts of Campomanesia adamantium leaves and fruits were evaluated against prostate cancer cells. In vitro studies showed that the isolated chalcone cardamonin inhibited prostate cancer cell proliferation and decreased the expression of NFkB1. [Molecules] Abstract | Full Article

Methylseleninic Acid Elevates REDD1 and Inhibits Prostate Cancer Cell Growth Despite AKT Activation and mTOR Dysregulation in Hypoxia
In both PTEN+ and PTEN- prostate cancer cells researchers showed that methylseleninic acid elevates REDD1 and phosphorylation of AKT along with p70S6K in hypoxia. [Cancer Med] Full Article

    CLINICAL RESEARCH

ERG Rearrangement Is Associated with Prostate Cancer-Related Death in Chinese Prostate Cancer Patients
Using fluorescence in situ hybridization and immunohistochemistry, researchers evaluated ETS-related gene (ERG), phosphatase tensin homologue and EGFR family aberrations in a cohort of 224 Chinese prostate cancer patients diagnosed in transurethral resection of the prostate. [PLoS One] Full Article

Treatment Outcomes in Very High-Risk Prostate Cancer Treated by Dose-Escalated and Combined-Modality Radiation Therapy
Scientists conducted a retrospective multi-institutional review on patients with very high-risk prostate cancer, treated with either dose-escalated external-beam radiotherapy or combined-modality radiotherapy consisting of pelvic irradiation and permanent interstitial brachytherapy. [Am J Clin Oncol] Abstract
