New Role for Prostate Cancer Protein Could Lead to Better Diagnosis and Treatment
Scientists studying how prostate cancer cells grow say they’ve discovered a new role for a protein previously known to be involved in repairing damaged DNA. The protein DNA-PK has now been shown to be involved in regulating the activity of genes involved in cancer spread. Effect of Extracellular Calcium on Regucalcin Expression and Cell Viability in Neoplastic and Non-Neoplastic Human Prostate Cells
Researchers aimed to unveil the role of extracellular calcium (Ca2+0) on regucalcin (RGN) expression and viability of non-neoplastic and neoplastic prostate cell lines. They demonstrated that Ca2+0 up-regulates RGN expression in both cell lines, but important differences were found between cells for dose- and time-responses to Ca2+0 treatment. [Biochim Biophys Acta] Abstract

Regulation of Human UDP-Glucuronosyltransferase 2B15 and 2B17 by miR-376c in Prostate Cancer Cell Lines
Scientists identified a putative miR-376c target site in the 3′-untranslated regions of both UGT2B15 and UGT2B17 mRNAs. In accord with the possibility that this miRNA negatively regulates UGT2B15 and UGT2B17 expression, there is an inverse correlation in the levels of miR-376c and UGT2B15/UGT2B17 mRNAs in prostate cancer cell lines versus normal prostate tissue. [J Pharmacol Exp Ther] Abstract

Characterization of Autoimmune Inflammation Induced Prostate Stem Cell Expansion
Ovalbumin specific CD8+ T cells were intravenously delivered to intact and castrated prostate ovalbumin expressing transgenic-3 mice to induce inflammation. Lin (CD45/CD31)Sca1+CD49f+ cells (LSC) and progenitor cells within LSC were determined by flow cytometry. Sorted LSC were subjected to a prostate sphere forming assay to evaluate prostate stem cell clonal propagation, proliferation, immediate differentiation, and self-renewal ability. Repurposing of Phentolamine as a Potential Anticancer Agent against Human Castration-Resistant Prostate Cancer: A Central Role on Microtubule Stabilization and Mitochondrial Apoptosis Pathway
Phentolamine induced anti-proliferative effect in PC-3 and DU-145, two castration-resistant prostate cancer cell lines, and P-glycoprotein overexpressing cells. This effect was not significantly reduced in paclitaxel-resistant cells. [Prostate] Abstract

Poly (Archive) Binding Protein Cytoplasmic 1 Is a Novel Co-Regulator of the Androgen Receptor
Knockdown of poly (Archive) binding protein cytoplasmic 1 decreased nuclear androgen receptor (AR) protein levels and inhibited androgen activation of the AR target PSA in LNCaP and C4-2 cells. [PLoS One] Full Article

P2Y2 Receptor and EGFR Cooperate to Promote Prostate Cancer Cell Invasion via ERK1/2 Pathway
Investigators found that extracellular ATP/UTP induced activation of EGFR and ERK1/2. Knockdown of IL-8 inhibited ATP-mediated invasion and migration of prostate cancer cells. [PLoS One] Full Article

Effects of MicroRNA-221/222 on Cell Proliferation and Apoptosis in Prostate Cancer Cells
Researchers investigated the role of miR-221/222 in cell proliferation and apoptosis in human prostate cancer cells, and examine the effects of miR-221/222 on caspase-10 expression. [Gene] Abstract

    CLINICAL RESEARCH

The Incidence and Relative Risk of Cardiovascular Toxicity in Patients Treated with New Hormonal Agents for Castration-Resistant Prostate Cancer
New hormonal agents are available for treating metastatic castration-resistant prostate cancer (mCRPC). The authors aimed to define the incidence and relative risk of cardiovascular events in mCRPC patients treated with these agents. [Eur J Cancer] Abstract

A Randomized, Double-Blind, Placebo-Controlled Study of the Effects of Pomegranate Extract on Rising PSA Levels in Men following Primary Therapy for Prostate Cancer
The authors compared the effects of pomegranate juice on PSA doubling times in subjects with rising PSA levels after primary therapy for prostate cancer. [Prostate Cancer Prostatic Dis] Abstract

Metabolomic Profiling for the Identification of Novel Diagnostic Markers in Prostate Cancer
During neoplastic transformation, prostate cells undergo metabolic reprogramming to satisfy the demands of growth and proliferation. The role of emerging biomarkers is summarized, based on the current understanding of the prostate cancer metabolome. [Expert Rev Mol Diagn] Abstract
