Genomic Hallmarks of Localized, Non-Indolent Prostate Cancer

Prostate tumors are highly variable in their response to therapies, but clinically available prognostic factors can explain only a fraction of this heterogeneity. Researchers analyzed 200 whole-genome sequences and 277 additional whole-exome sequences from localized, non-indolent prostate tumors with similar clinical risk profiles, and carried out RNA and methylation analyses in a subset. Targeting Plk1 to Enhance Efficacy of Olaparib in Castration-Resistant Prostate Cancer

Olaparib is a FDA-approved PARP inhibitor (PARPi) that has shown promise as a synthetic lethal treatment approach for BRCA-mutant castration-resistant prostate cancer (CRPC) in clinical use. However, emerging data has also shown that even BRCA-mutant cells may be resistant to PARPi. Herein, the authors showed a robust sensitization of Olaparib in 22RV1, a BRCA1 deficient CRPC cell line, as well as in CRPC xenograft tumors. [Mol Cancer Ther] Abstract

Metabolic Profiling in Formalin-Fixed and Paraffin Embedded Prostate Cancer Tissues

Mass spectrometry-based metabolomic analysis was performed in matched frozen and formalin-fixed paraffin-embedded human prostate cancers as well as isogenic prostate cancer cell lines. Sensitization of Radioresistant Prostate Cancer Cells by Resveratrol Isolated from Arachis hypogaea Stems

The authors characterized resveratrol (RV, 3,4ʹ,5-trihydroxystilbene) in peanut stem extract (PSE) for the first time and showed that both RV and PSE dose-dependently induced cell death in DOC-2/DAB2 interactive protein-deficient prostate cancer cells with the radioresistant phenotype. Knockdown of PYCR1 Inhibits Cell Proliferation and Colony Formation via Cell Cycle Arrest and Apoptosis in Prostate Cancer
