Asporin Is a Stromally Expressed Marker Associated with Prostate Cancer Progression

Researchers used Kaplan–Meier, univariate and multivariate analysis to correlate the expression of Asporin (ASPN) mRNA and protein with prostate cancer progression in independent cohorts. They used cell cultures of primary prostate cancer fibroblasts treated with serum-free conditioned media from prostate cancer cell lines to examine regulation of ASPN mRNA in tumor stromal cells. Proteasome-Associated Deubiquitinase Ubiquitin-Specific Protease 14 Regulates Prostate Cancer Proliferation by Deubiquitinating and Stabilizing Androgen Receptor

The authors identified ubiquitin-specific protease 14 as a novel regulator of androgen receptor (AR), inhibiting the degradation of AR via deubiquitinating this oncoprotein in the androgen-responsive prostate cancer cells. [Cell Death Dis] Full Article

The Role of Epithelial–Mesenchymal Transition Drivers ZEB1 and ZEB2 in Mediating Docetaxel-Resistant Prostate Cancer

Scientists characterized epithelial–mesenchymal transition (EMT) in docetaxel-resistant sublines through increased invasion, MMP-1 production and ZEB1 and ZEB2 expression. They presented evidence for differential EMT across PC-3 and DU145 in vitro resistance models as characterized by differential migration, cell colony scattering and susceptibility to the cancer stem cell inhibitor salinomycin. Targeted Nanoparticles Encapsulating (−)-Epigallocatechin-3-Gallate for Prostate Cancer Prevention and Therapy

Investigators developed polymeric epigallocatechin-3-gallate-encapsulated nanoparticles targeted with small molecular entities, able to bind to prostate specific membrane antigen, a transmembrane protein that is overexpressed in prostate cancer, and evaluated their efficacy in preclinical studies. Litchi Seed Extracts Diminish Prostate Cancer Progression via Induction of Apoptosis and Attenuation of EMT through Akt/GSK-3β Signaling

Scientists used n-butyl alcohol extract of Litchi seed (NLS) to treat prostate cancer PC3, DU145, RM1 and C4-2B cells. NLS induced a significant decrease in cell viability and clonogenic growth in a dose-dependent manner. [Sci Rep] Full Article

CK2 Abrogates the Inhibitory Effects of PRH/HHEX on Prostate Cancer Cell Migration and Invasion and Acts through PRH to Control Cell Proliferation

The authors showed that proline-rich homeodomain protein (PRH) and hyper-phosphorylated PRH are present in normal prostate epithelial cells, and that hyper-phosphorylated PRH levels are elevated in benign prostatic hyperplasia, prostatic adenocarcinoma, and prostate cancer cell lines. Minnelide Inhibits Androgen Dependent, Castration Resistant Prostate Cancer Growth by Decreasing Expression of Androgen Receptor Full Length and Splice Variants

Researchers synthesized a water-soluble pro-drug of triptolide, minnelide, that is currently being evaluated in a Phase I clinical trial against gastrointestinal tumors. In the current study, they assessed the therapeutic potential of minnelide and its active compound triptolide against androgen dependent prostate cancer both in vitro as well as in vivo. Distinct Prostate Cancer-Related mRNA Cargo in Extracellular Vesicle Subsets from Prostate Cell Lines

Scientists hypothesized that the messenger RNA (mRNA) cargo of extracellular vesicles (EVs) could differ based on the EV cellular origin and subpopulation analyzed. They isolated microvesicles and exosomes from PC-3 and LNCaP prostate cancer cells by differential centrifugation and compared them to EVs derived from the benign PNT2 prostate cells.
