TOP STORYResearchers found that speckle-type POZ protein (SPOP)WT can physically interact with c-MYC protein and, upon exogenous expression in vitro, can promote c-MYC ubiquitination and degradation. This effect was attenuated in prostate adenocarcinoma (PC) cells by introducing PC-associated SPOP mutants or upon knockdown of SPOP via short-hairpin-RNA. [Oncogene] Abstract | |

PUBLICATIONS(Ranked by impact factor of the journal)LABORATORY RESEARCHScientists showed that monoamine oxidase A (MAOA) is involved in mediating neuroendocrine differentiation of prostate cancer (PCa) cells, a feature associated with hormone-refractory PCa, a lethal type of disease. [Sci Rep] Full Article Investigators showed that transcriptional co-activator with PDZ-binding motif (TAZ) is a basal cell marker for the prostate epithelium. They found that overexpression of TAZ promoted the epithelial-mesenchymal transition, cell migration, and anchorage-independent growth in the RWPE1 prostate epithelial cells. [J Biol Chem] Abstract Scientists found for the first time that isorhapontigenin apparently induced cell growth inhibition and apoptosis by targeting EGFR and its downstream signal pathways in prostate cancer cells both in vitro and in vivo, whereas no obviously effect on normal prostate cells. [J Cell Physiol] Abstract Researchers showed that androgen could induce a quiescent state that is self-sustained in a cell-autonomous manner through a “hit and run” mechanism in androgen receptor-expressing prostate cancer cells. [Cell Cycle] Abstract GABABR-Induced EGFR Transactivation Promotes Migration of Human Prostate Cancer Cells Scientists investigated whether metabotropic GABAB receptor (GABABR) hijacks receptor tyrosine kinase (RTK) signaling to modulate the fates of human prostate cancer cells. RTKs array analysis revealed that GABABR specific agonist, baclofen, selectively induced the transactivation of EGFR in PC-3 cells. [Mol Pharmacol] Abstract The authors identified glycogen synthase kinase-3 inhibitors as transcriptional suppressors of androgen receptor (AR)-V7 using a high-throughput screen with an AR-V7 reporter system. LY-2090314 suppressed the reporter activity and endogenous AR-V7 activity in JDCaP-hr cells. [Prostate] Abstract Androgen Receptor-Regulated miRNA-193a-3p Targets AJUBA to Promote Prostate Cancer Cell Migration To investigate the role of miRNAs in prostate cancer progression, researchers performed small RNA-sequencing analysis in androgen-dependent LNCaP cells and LNCaP-derived castration-resistant prostate cancer C4-2B cells. For functional validation, they specifically investigated miR-193a-3p, which is highly upregulated in C4-2B cells and modulated by the androgen receptor. [Prostate] Abstract Investigators developed a lentiviral vector, PLKO.1-sh specific cysteine protease 1 (SENP1), to silence SENP1 in prostate cancer cells with high metastatic characteristics (PC3M). Likewise, they also created an adenovirus vector, Ad5/F11p-SENP1 to over-express SENP1 in prostate cancer cells with low metastatic potential. They showed that silencing of SENP1 promoted cellular apoptosis, and inhibited proliferation and migration of PC3M cells. [Int J Mol Sci] Full Article CLINICAL RESEARCHThe authors investigated whether the addition of rhenium-188-HEDP to docetaxel/prednisone improved efficacy of chemotherapy in patients with CRPC. [Eur J Nucl Med Mol Imaging] Abstract This Phase I dose escalation study of UV1 evaluated safety, immune response, effects on prostate-specific antigen levels, and preliminary clinical outcome. REVIEWSCellular Determinants and Microenvironmental Regulation of Prostate Cancer Metastasis The authors provide a general summary on cancer metastasis and a specific discussion on prostate cancer (PCa) metastasis. They discuss recent advances in the knowledge of the cellular determinants of PCa metastasis and the importance of tumor microenvironment, especially an immunosuppressive tumor microenvironment, in shaping metastatic propensities. INDUSTRY NEWSCellular Dynamics International Signs Distribution Deal with STEMCELL Technologies Cellular Dynamics International announced it has signed a distribution agreement with STEMCELL Technologies, a world leader in iPS cell culture media. [STEMCELL Technologies Inc.] Press Release UCLA Researcher Awarded Prestigious Grant to Study Role of Molecule in Prostate Cancer UCLA researcher Dr. Andrew Goldstein has been awarded a research scholar grant from the American Cancer Society to fund his work on prostate cancer. Goldstein’s grant will support his efforts to define the role of a molecule called CD38, which may play a key role in preventing aggressive prostate cancers. POLICY NEWSPower Struggle Erupts at Utah Cancer Institute over Director’s Firing The abrupt dismissal of the head of a Utah cancer center is causing backlash from its faculty—and its major philanthropic funder—in a struggle over the center’s autonomy from the University of Utah in Salt Lake City. And nearly 2000 researchers have signed a petition calling on the university to reverse its decision. [ScienceInsider] Editorial For Congress, March for Science Is a Democratic Event Organizers of the March for Science have deliberately avoided reaching out to U.S. elected officials, saying that they want the rallies to be apolitical. As a result, few members of Congress will be participating in the main event in Washington, D.C., and at hundreds of satellite marches across the country. [ScienceInsider] Editorial
