TOP STORYDifferential Impact of RB Status on E2F1 Reprogramming in Human Cancer Analysis of tumor samples and cell-free DNA from patients with advanced prostate cancer showed that direct retinoblastoma (RB) loss was the preferred pathway of disruption in human disease. While RB loss was associated with lethal disease, RB-deficient tumors had no proliferative advantage and exhibited downstream effects distinct from cell cycle control. PUBLICATIONS(Ranked by impact factor of the journal)Preclinical studies using multiple in vitro prostate cancer (PCa) cell lines and an in vivo mouse model with xenografted castration-resistant PCa CWR22Rv1 cells demonstrated that ASC-J9® has similar AR degradation effects when dissolved in FDA-approved solvents, including DMSO, PEG-400:Tween-80, DMA:Labrasol:Tween-80, and DMA:Labrasol:Tween-20. [Cancer Lett] Abstract miR-141-3p expression was reduced in bone metastatic prostate cancer (PCa) tissues compared with non-bone metastatic PCa tissues. Low expression of miR-141-3p positively correlated with serum PSA levels, Gleason grade and bone metastasis status in PCa patients. Furthermore, upregulating miR-141-3p suppressed the epithelial–mesenchymal transition, invasion and migration of PCa cells in vitro. [J Exp Clin Cancer Res] Full Article Scientists undertook RNA sequencing analysis of prostate tumors and adjacent non-malignant cells to identify polymorphic short tandem repeats that are readily expressed in these cells. [Sci Rep] Full Article The authors focused on 20β-hydroxy-5α-dihydroprogesterone (20β-OHDHP), a C21s metabolized from progestin, and showed that 20β-OHDHP is synthesized in prostate cancer cells and is able to directly stimulate the androgen receptor. [Prostate] Abstract LNCaP and VCaP growth was stimulated when cells were cultured in serum free media (SFM) or stripped fetal bovine serum mediam (CSM) supplemented with testosterone (T). LNCaP, C4-2, LAPC-4, and VCaP cell growth was inhibited when cultured in SFM or CSM with T and bicalutamide. [Prostate] Abstract Investigators found that CDKN3 was highly expressed in prostate cancer and negatively correlated with disease relapse. CDKN3 positively controlled the cell proliferation in prostate carcinoma cell lines. [Biomed Pharmacother] Abstract Hypermethylation of the SPARC Promoter and Its Prognostic Value for Prostate Cancer Scientists investigated the abnormalities in the expression of secreted protein acidic and rich in cysteine (SPARC) and its promoter hypermethylation in prostate cancers and in correlated clinicopathological profiles. [Oncol Rep] Abstract MiR-199a Suppresses Prostate Cancer Paclitaxel Resistance by Targeting YES1 The downregulation of miR-199a contributes to paclitaxel (PTX) resistance in prostate cancer. Yamaguchi sarcoma viral homolog 1 (YES1) mediated the regulation of miR-199a in prostate cancer PTX resistance. [World J Urol] Abstract Prohibitin-2 Negatively Regulates AKT2 Expression to Promote Prostate Cancer Cell Migration Researchers induced overexpression and knockdown of PHB2 in prostate cancer (PCa) cell lines with the aim of examining the effects of prohibitin-2 on PCa cell migration via wound healing assays. REVIEWSCross-Talk between Protein Synthesis, Energy Metabolism and Autophagy in Cancer The authors discuss emerging findings that highlight hitherto unappreciated aspects of signaling to the translation apparatus with the particular focus on emerging connections between protein synthesis, autophagy and energy homeostasis in cancer. [Curr Opin Genet Dev] Abstract Scientists provide the most updated information on the recent methods of prostate cancer screening, imaging and treatment modalities. Wherever possible, clinical trial data has been supplemented to provide a comprehensive overview of current prostate cancer research and development. POLICY NEWSBrexit “Breakthrough” Draws Praise from Science Policy Experts In laboratories throughout the UK, researchers and administrators gave out a collective sigh of relief following a decision to allow EU nationals to remain after the country leaves the union in March 2019. [Nature News] Editorial U.S. Science Groups Make Last-Minute Push to Influence Final Tax Deal It wasn’t your typical graduate school experience. Eight graduate students, including doctoral students in anthropology and astrophysics at major U.S. universities, ended up in handcuffs after they refused to end a noisy protest outside the Washington, D.C., office of Speaker of the House of Representatives Paul Ryan. [ScienceInsider] Editorial US Graduate Students in Uproar over Proposed Tax Hike Many US universities waive tuition for students who conduct research or teach. But a provision in a tax bill passed by the US House of Representatives would add that tuition to students’ taxable income. [Nature News] Editorial
