YAP1 and AR Interactions Contribute to the Switch from Androgen-Dependent to Castration-Resistant Growth in Prostate Cancer
Scientists identified yes-associated protein 1 (YAP1), as a physiological binding partner and positive regulator of androgen receptor (AR) in prostate cancer. YAP1 and AR co-localize and interact with each other predominantly within cell nuclei by an androgen-dependent mechanism in a hormone naive and an androgen-independent mechanism in castration-resistant prostate cancer cells. The Inhibitory Effects of AR/miR-190a/YB-1 Negative Feedback Loop on Prostate Cancer and Underlying Mechanism
Researchers observed androgen receptor (AR)/miR-190a/YB-1 forms an auto-regulatory negative feedback loop in prostate cancer: miR-190a expression was down-regulated by AR activation; YB-1 functions are as an AR activator; miR-190a inhibited AR expression and transactivation through direct binding to 3′UTR of YB-1 gene. A Novel Recombinant Slow-Release TNF α-Derived Peptide Effectively Inhibits Tumor Growth and Angiogenesis
RMP16 had significant inhibition effects for multiple tumor cells, especially prostate cancer Du145 cells, and human vascular endothelial cells but not for human mammary non-tumorigenic epithelial cells. [Sci Rep] Full Article

MicroRNA Expression Signature of Castration-Resistant Prostate Cancer: The MicroRNA-221/222 Cluster Functions as a Tumor Suppressor and Disease Progression Marker
Researchers investigated the functional roles of microRNA (miRNA)-221 and miRNA-222 in prostate cancer cells. Restoration of miR-221 or miR-222 in cancer cells revealed that both miRNAs significantly inhibited cancer cell migration and invasion. [Br J Cancer] Abstract

ATM Inhibition Potentiates Death of Androgen Receptor-Inactivated Prostate Cancer Cells with Telomere Dysfunction
Since telomere dysfunction in other systems is known to activate ATM (ataxia telangiectasia mutated)-mediated DNA damage response (DDR) signaling pathways, scientists investigated the role of ATM-mediated DDR signaling in androgen receptor-inactivated prostate cancer cells. [J Biol Chem] Abstract | Full Article

Anti-Tumor Effect of the Alphavirus-Based Virus-Like Particle Vector Expressing Prostate-Specific Antigen in a HLA–DR Transgenic Mouse Model of Prostate Cancer
Researchers determined if an alphavirus-based vaccine encoding human prostate-specific antigen (PSA) could generate an effective anti-tumor immune response in a stringent mouse model of prostate cancer. They found that virus-like particle vector encoding PSA could infect mouse dendritic cells in vitro and induce a robust PSA-specific immune response in vivo. [Vaccine] Abstract

Cyanidin Induces Apoptosis and Differentiation in Prostate Cancer Cells
Researchers delineated the anti-proliferative activity and the cytodifferentiation properties mediated by cyanidin-3-O-β-glucopyranoside (C3G) treatment in the DU145 and LnCap human prostatic cancer cell lines. C3G produced anti-proliferative effects through activation of caspase-3 and induction of p21 protein expression. [Int J Oncol] Abstract

The Resveratrol Tetramer r-Viniferin Induces a Cell Cycle Arrest Followed by Apoptosis in the Prostate Cancer Cell Line LNCaP
Scientists investigated the cytotoxic/growth-inhibiting properties of the resveratrol tetramer r-viniferin on the prostate cancer cell line LNCaP and compared them with those of resveratrol. [Phytother Res] Abstract

Cardiolipin Composition Correlates with Prostate Cancer Cell Proliferation
Investigators analyzed the interrelationship between cardiolipin (CL) composition and the proliferation of prostate cells by exposing PC-3 tumor cells to different fatty acids and by analyzing the CL composition in prostate tissue from prostate cancer patients after prostatectomy. [Mol Cell Biochem] Abstract

Comparison of Two Adjuvant Hormone Therapy Regimens in Patients with High-Risk Localized Prostate Cancer after Radical Prostatectomy: Primary Results of Study CU1005
Researchers evaluated two regimens of maximum androgen blockage or bicalutamide 150 mg daily as immediate adjuvant therapy for high-risk localized prostate cancer. [Asian J Androl] Full Article

The Many Ways to Make a Luminal Cell and a Prostate Cancer Cell
Given that there are multiple cells that can regenerate prostatic tissue and give rise to prostate cancer, the focus should shift to defining the signaling mechanisms that drive differentiation and progenitor self-renewal. The authors review the literature, present the evidence and raise important unanswered questions that will help guide the field forward to dissecting critical mechanisms regulating stem cell differentiation and tumor-initiation. INDUSTRY NEWS
Quebec Researchers to Fight a Deadly Form of Children’s Leukemia and to Test a New Way to Kill Prostate Cancer Cells
Two of three Quebec researchers to receive funding from the Terry Fox Research Institute will pursue new ways to help children in hospital who are battling a deadly form of leukemia. A third-funded researcher will test a recent molecular finding to see how the immune system might help kill prostate cancer tumor cells. [The Terry Fox Research Institute] Press Release

Exploring the Use of Aspirin and a Diabetes Drug for Prostate Cancer Prevention and Control
Rutgers Cancer Institute of New Jersey population science researcher XiangLin Tan, MD, PhD, has been awarded an $852,705 Career Development Award from the National Cancer Institute to explore the use of aspirin and a diabetes drug for prostate cancer prevention and control, especially in obese men. NCI-Designated Cancer Centers Expand, Four Awarded Comprehensive Designation
With the addition of the Tisch Cancer Institute at the Icahn School of Medicine at Mount Sinai in New York City, National Cancer Institute’s (NCI)’s cancer centers program now has 69 cancer centers in 35 states. The upgraded designations for the four centers that were already part of the NCI program—from standard cancer centers to comprehensive cancer centers—brings the total number of comprehensive cancer centers to 45.
